呼吸道合胞病毒感染性肺炎患儿的TNF-α和IL-1β水平及TLR4信号通路作用初步探究 被引量:34

Preliminary study on levels of TNF-α and IL-1β, and role of TLR4 signaling pathway in children with respiratory syncytial virus infectious pneumonia

摘要 目的初步探讨呼吸道合胞病毒(RSV)感染性肺炎患儿的肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)水平及Toll样受体4(TLR4)信号通路作用。方法选择攀枝花市第三人民医院于2017年1月-2019年1月收治的RSV感染性肺炎患儿80例作为病例组;选择医院同期收治的普通肺炎患儿60例作为普通肺炎组,健康体检小儿50名作为对照组。采用酶联免疫吸附法测定TNF-α和IL-1β含量;采用流式细胞仪检测单核细胞TLR4蛋白阳性率;采用Real-time PCR检测TLR4信号通路表达。结果相比普通肺炎组和对照组,病例组血清TNF-α和IL-1β水平升高,且普通肺炎组高于对照组(P<0.05)。相比普通肺炎组和对照组,病例组单核细胞TLR4 mRNA和A20 mRNA表达升高,且普通肺炎组高于对照组(P<0.05)。相比普通肺炎组和对照组,病例组单核细胞TLR4蛋白表达率升高,且普通肺炎组高于对照组(P<0.05)。结论 RSV感染性肺炎患儿血清TNF-α和IL-1β水平高表达,单核细胞TLR4信号通路可能参与了RSV感染性肺炎的炎症反应。 OBJECTIVE To preliminarily explore the levels of tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β) and the role of Toll-like receptor 4(TLR4) signaling pathway in children with respiratory syncytial virus(RSV) infectious pneumonia. METHODS Totally 80 children with RSV infectious pneumonia who were treated in the Third People’s Hospital of Panzhihua from Jan 2017 to Jan 2019 were chosen as the case group, meanwhile, 60 children with common pneumonia were set as the common pneumonia group, and 50 healthy children who received physical examination were assigned as the control group. The levels of TNF-α and IL-1β were determined by using enzyme-linked immunosorbent assay, the positive rate of TLR4 protein in monocytes was detected with the use of flow cytometry, and the expression of TLR4 signaling pathway was detected by means of real-time PCR. RESULTS The levels of serum TNF-α and IL-1β of the case group were significantly higher than those of the common pneumonia group and the control group, and the levels of above indexes of the common pneumonia group were significantly higher than those of the control group(P<0.05). The expression levels of TLR4 mRNA and A20 mRNA in monocytes were significantly higher in the case group than in the common pneumonia group and the control group, and the expression levels of above indexes were significantly higher in the common pneumonia group than in the control group(P<0.05). The positive rate of TLR4 protein in monocytes of the case group was significantly higher than that of the common pneumonia group and the control group, and the positive rate of TLR4 protein in monocytes of the common pneumonia group was significantly higher than that of the control group(P<0.05). CONCLUSION The children with RSV infectious pneumonia show high expression levels of serum TNF-α and IL-1β, and the TLR4 signaling pathway in monocytes may be involved in inflammatory response of RSV infectious pneumonia.
