摘要
目的研究山茱萸生品及不同炮制品分别配伍入方的六味地黄汤对绝经后骨质疏松症的干预作用及其机制。方法将80只雌性SD大鼠随机分为空白组(生理盐水,10 mL·kg^-1),假手术组(生理盐水,10 mL·kg^-1),模型组(生理盐水,10 mL·kg^-1)以及山茱萸生品(生茱萸)、山茱萸酒制品(酒茱萸)、山茱萸蒸制品(蒸茱萸)、山茱萸盐制品(盐茱萸)分别配伍入方的六味地黄汤生茱萸组(生茱萸组,6.75 g·kg^-1)、六味地黄汤酒茱萸组(酒茱萸组,6.75 g·kg^-1)、六味地黄汤盐茱萸组(盐茱萸组,6.75 g·kg^-1)、六味地黄汤蒸茱萸组(蒸茱萸组,6.75 g·kg^-1),阿伦磷酸钠对照组(阳性药物组,6.3 mg·kg^-1),每组10只。除空白组外,其余组均采用切除雌性SD大鼠双侧卵巢的方式建立绝经后骨质疏松症模型。造模后第5日开始,各组按相应剂量连续灌胃给药90 d,每日一次。90 d后用双能X线骨密度仪、ELISA法及放射免疫法检测大鼠股骨骨密度(BMD)值、血钙及血磷的含量;碱性磷酸酶(ALP)、抗酒石酸酸性磷酸酶(TRAP)、血清骨钙素(BGP)的水平。结果与模型组比较,生茱萸组、酒茱萸组、盐茱萸组、蒸茱萸组的六味地黄汤均可以显著提高绝经后骨质疏松症模型大鼠的BMD值及血磷含量,降低ALP的水平(P<0.05),生茱萸组、酒茱萸组、蒸茱萸组的六味地黄汤可以降低绝经后骨质疏松症模型大鼠TRAP水平(P<0.05);酒茱萸组六味地黄汤还可提高血钙含量(P<0.05),酒茱萸、盐茱萸组六味地黄汤可以降低BGP水平(P<0.05)。结论山茱萸生品及其不同炮制品分别配伍入方的六味地黄汤均可改善绝经后骨质疏松症,其中对骨密度的改善作用以酒茱萸组为优,且炮制品组优于生茱萸组;酒茱萸组、盐茱萸组、蒸茱萸组六味地黄汤的抗绝经后骨质疏松症作用没有显著差异,临床可以根据具体情况选择合适的炮制品配伍入六味地黄汤方防治绝经后骨质疏松症。
Objective To study the effect and mechanism of Liuwei Dihuang decoction with different processed Corni Fructus on ovariectomized postmenopausal osteoporosis model rats. Methods Eighty female SpragueDawley rats were randomly divided into a blank group(normal saline, 10 mL·kg^-1), a sham operation group(normal saline, 10 mL·kg^-1), a model group(normal saline, 10 mL·kg^-1), and Corni Fructus raw products(raw Corni Fructus), Corni Fructus with wine(wine Corni Fructus), Corni Fructus processed by steam(steamed Corni Fructus), Corni Fructus with salt(salt Corni Fructus), in combination with Liuwei Dihuang decoction(raw Corni Fructus group, 6.75 g·kg^-1), Liuwei Dihuang decoction wine Corni Fructus Group(wine Corni Fructus group, 6.75 g·kg^-1), Liuwei Dihuang decoction salt Corni Fructus group(salt Corni Fructus group, 6.75 g·kg^-1), Liuwei Dihuang decoction steamed Corni Fructus group(steamed Corni Fructus group, 6.75 g·kg^-1), and an alendronate sodium control group(positive drug group, 6.3 mg·kg^-1), 10 in each group. Except for the blank group, the postmenopausal osteoporosis models were established by excising the bilateral ovaries in female SD rats. On the 5 th day after the model establishment, each group was continuously intragastrically administered for 90 days, once daily. After 90 days, the femoral bone mineral density(BMD), blood calcium(Ca) and blood phosphorus(P) content, alkaline phosphatase(ALP), tartrate-resistant acid phosphatase(TRAP), and serum osteocalcin(BGP) levels were detected by dual energy X-ray absorptionmetry, ELISA, and radioimmunoassay. Results Compared with the model group, Liuwei Dihuang decoction can significantly improved the BMD and blood phosphorus content in the model rats with postmenopausal osteoporosis, and reduced the levels of ALP and TRAP(P < 0.05) in all groups. Liuwei Dihuang decoction in the wine Corni Fructus group also increased the blood calcium content(P < 0.05) and decreased BGP level(P < 0.05). Conclusion Liuwei Dihuang decoction compatible with Corni Fructus
