摘要
目的:研究不同负荷运动对慢性心力衰竭大鼠心功能及心肌细胞自噬相关蛋白表达的影响,探讨运动训练对心力衰竭后心肌的保护机制。方法:采用健康SD大鼠40只,左冠状动脉前降支(LAD)结扎法制作心肌梗死后心力衰竭模型。术后4周,12导联心电图检测有6~8个Q波、且超声心动检查左心室射血分数(EF)30%~50%的模型大鼠入选后续实验。共有32只大鼠入选,随机分为模型对照组(C+LAD组)、模型低强度运动训练组(LE+LAD组)、模型中等强度运动训练组(ME+LAD组)、模型高强度运动训练组(HE+LAD组),每组8只,分别进行不同强度运动训练。训练8周后,应用超声心动和Western-blot法分别检测模型各组大鼠的心功能和心肌细胞自噬相关蛋白雷帕霉素靶蛋白(mTOR)、磷酸化雷帕霉素靶蛋白(p-mTOR)、自噬相关蛋白LC3B的表达情况。结果:与C+LAD组比较,不同强度运动训练组大鼠左心室射血分数(EF)和短轴缩短率(FS)值均有不同程度升高,以ME+LAD组升高程度最为显著(P<0.01)。与LE+LAD组相比,ME+LAD组和HE+LAD组大鼠心肌组织p-mTOR表达降低、LC3B-Ⅱ表达升高(P<0.01)。结论:不同负荷运动训练能够对心力衰竭大鼠心功能和心肌自噬相关蛋白表达产生不同程度影响。适宜强度运动训练对心功能的改善作用可能通过调节心肌细胞自噬实现。
Objective To investigate the effect of exercises of different intensitieson the cardiac function and expression of autophagy-related proteins in cardiomuyocytes of rats with chronic heart failure(CHF),and to explore the protective mechanism of the exercise training on the myocardium after heart failure.Methods Forty healthy Sprague-Dawley rats were chosen and established the CHF model using the left anterior descending coronary artery(LAD) ligation.Four weeks after the surgery,32 rats with six to eight lead Q waves in the 12-lead electrocardiogram and 30% to 50% left ventricular ejection fraction(EF) values were selected into the following study,and randomly divided into four groups according to their EF values:a control group(C+LAD),a low-intensity exercise group(LE+LAD),a moderate-intensity exercise group(ME+LAD) and a high-intensity exercise group(HE+LAD),each of 8.They underwent exercises of different intensities as their group names implied.After an eight-week training,the heart function and expression of autophagy-related proteins including mammalian target of rapamycin(mTOR),phosphorylation mTOR(p-mTOR) and microtubule-associated protein light chain 3 B(LC3 B) were detected using the echocardiography and Western blotting.Results Compared with C+LAD group,significant improvement was observed in the average EF and fractional shortening values in the other groups,with those of ME+LAD group most significant(P<0.01).The expression of p-mTOR of ME+LAD and HE+LAD groups was lower than LE+LAD,while that of LC3 B-Ⅱwas higher than the latter group(P<0.01).Conclusion Exercises at different intensities have different effect on the cardiac function and expression of autophagy-related proteins in cardiomyocytes of CHF rats.Exercises at an appropriate intensity may improve the heart function through regulating the autophagy of cardiomyocytes.
