摘要
目的探讨良、恶性宫颈癌及正常宫颈组织中自噬相关基因Beclin1和Atg5的表达情况,分析其在宫颈癌病程发生发展中的作用。方法将广东省廉江市人民医院收治的91例患者分为宫颈癌组、上皮内瘤变组和正常宫颈组,分别取各组宫颈组织并通过western blot、免疫组化及PCR法检测Beclin1和Atg5的表达情况。结果Beclin1和Atg5在宫颈癌组中mRNA和蛋白表达水平均显著低于正常宫颈组和上皮内瘤变组。结论宫颈癌组织中Beclin1和Atg5表达较正常宫颈组织显著下降,提示宫颈癌的发生发展可能与自噬相关基因Beclin1和Atg5的活性降低有关。
Objective To investigate the expression of autophagy-related genes Beclin1 and Atg5 in benign and malignant cervical cancer and normal cervical tissues,and to analyze their role in the pathogenesis and development of cervical cancer.Methods 91 patients admitted to our hospital were divided into cervical cancer group,intraepithelial neoplasia group and normal cervical group.The cervical tissues of each group were taken and the expression of Beclin1 and Atg5 was detected by western blot,immunohistochemistry and PCR.Results The mRNA and protein expression levels of Beclin1 and Atg5 in cervical cancer group were significantly lower than those in normal cervical and intraepithelial neoplasia groups.Conclusion The expression of Beclin1 and Atg5 in cervical cancer tissues was significantly lower than that in normal cervical tissues,suggesting that the development of cervical cancer may be related to the decreased activity of autophagy-related genes Beclin1 and Atg5.
