摘要
目的:建立前胡总香豆素提取物的质量标准。方法:采用超临界萃取法(SFE)提取前胡总香豆素类成分,薄层色谱法(TLC)、紫外分光光度法(UV)和高效液相色谱法(HPLC)等对前胡总香豆素提取物进行定性定量研究。结果:TLC斑点清晰,阴性无干扰;分别以白花前胡甲素和白花前胡乙素计,前胡总香豆素含量范围依次为78.69%~90.07%和85.62%~98.10%;特征图谱40 min内主要色谱峰分离情况良好,共标定5个特征峰并对2个主要特征峰含量进行测定,测得主要特征峰白花前胡甲素和白花前胡乙素的含量范围依次为602.42~721.44 mg/g和38.34~101.46 mg/g。结论:该方法精密、快速,专属性强,可用于前胡总香豆素提取物的质量控制。
Objective:To establish the quality standards for total coumarin extracts of Peucedani Radix.Methods:Supercritical fluid extraction(SFE)was used to extract the total coumarins from Peucedani Radix.Thin-layer chromatography(TLC),ultraviolet spectrophotometry(UV)and high performance liquid chromatography(HPLC)were used to study the qualitative and quantitative of total coumarin extracts.Results:The TLC spots were clearly observed without negative interference;Taking the Praeruptorin A and Praeruptorin B as reference,the total coumarin contents of Peucedani Radix were in the range of 78.69%~90.07%and 85.62%~98.10%.The main chromatograms of the characteristic map were completely separated within 40 minutes.Five characteristic peaks were marked and two characteristic peaks were determined.The contents of Praeruptorin A and the contents of Praeruptorin B were 602.42~721.44 mg/g and 38.34~101.46 mg/g respectively.Conclusion:The method is accurate,sensitive and specific,and can be used for the quality control of total coumarin extracts of Peucedani Radix.
