化学小分子靶标鉴定方法的研究进展

摘要 具有生物活性的化学小分子常通过与一个或多个细胞蛋白结合以调控细胞或组织的多种生物学效应,其靶标的鉴定成为完全了解药物作用机制的关键,也成为药物研发的重点内容。近年来在化学小分子靶标蛋白鉴定方面不断涌现出新技术和新方法,目前的鉴定方法是基于基因组学、蛋白质组学、克隆扩增、数据库比较分析、基于副作用报告系统的统计计算5个方面有效筛选并鉴定小分子化合物的靶标,各种方法均存在优势,但也存在局限性。随着研究的深入,更加完善的方法也将出现。 Bioactive small molecules often regulate biological effects by binding to one or more cellular proteins.The identification of their targets,which is the key to fully understand the mechanism of drug action,has become the focus of drug development.In recent years new technologies and methods are emerging in the identification of small chemical molecular targets.The existing screening tools are based on genomics,proteomics,clonal amplification comparative analysis of database and the statistical calculation of report system based on side effects to effectively screen and identify the target of the small molecular compound.Each method has its own advantages and limitations,and with the deepening of the research,better methods will also appear.
