补肺益肾方对肾精亏虚证大鼠睾丸支持细胞和生精功能的影响 被引量:5

Effect of Bufei Yishen Recipe on the Function of Sertoli Cells and Spermatogenesis of Testis in Rats with Deficiency of Shen Essence Syndrome

摘要 目的观察补肺益肾方对肾精亏虚证大鼠睾丸支持细胞和生精功能的影响。方法25只SD大鼠随机分为5组:正常组、模型组、五子衍宗方组、补肺益肾方低剂量组、补肺益肾方高剂量组,每组5只。正常组每天早晚给予5%羧甲基纤维素钠[CMC-Na,100 mg/(kg·d)],其余各组每早给予雷公藤多甙[GTW,20 mg/(kg·d)],模型组每晚给予5%CMC-Na,50 mg/(kg·d),五子衍宗方组每晚给予五子衍宗汤[15.8 g/(kg·d)],补肺益肾方低、高剂量组每晚给予补肺益肾方[6.6 g/(kg·d)、26.6 g/(kg·d)],均灌胃30天。计算脏器指数;采用Western Blot检测睾丸支持细胞波形蛋白(vimentin)、紧密连接蛋白11(claudin-11)和转铁蛋白(TF)表达;ELISA法测定血清抑制素B(INHB)水平;HE染色法和Johnsen评分观察并评价睾丸组织病理学变化。结果与正常组比较,模型组附睾指数、睾丸支持细胞vimentin、claudin-11、TF表达、INHB水平和Johnsen评分下降(P<0.05,P<0.01)。与模型组比较,五子衍宗方组和补肺益肾方高剂量组睾丸支持细胞vimentin、claudin-11、TF表达升高(P<0.05,P<0.01),各给药组INHB水平和Johnsen评分均升高(P<0.01)。与五子衍宗方组比较,补肺益肾方高剂量组睾丸支持细胞vimentin表达、INHB水平和Johnsen评分升高(P<0.05,P<0.01)。结论补肺益肾方高剂量可显著改善肾精亏虚证大鼠睾丸支持细胞和生精功能,其疗效优于五子衍宗方。 Objective To observe the effect of Bufei Yishen Recipe(BYR) on the function of sertoli cells and spermatogenesis in rats with deficiency of Shen essence syndrome. Methods Totally 25 SD rats were randomly divided into 5 groups, i.e., normal group, model group, Wuzi Yanzong Recipe(WYR) group, low-dose BYR group, and high-dose BYR group. The normal group was given 5% CMC-Na(100 mg/kg per day) every morning and evening, the other four groups were given tripterygium wilfordii(GTW, 20 mg/kg per day) every morning, the model group was given 5 % CMC-Na(50 mg/kg per day), WYR group were given WYR(15.8 g/kg per day), low-dose BYR group and high-dose BYR group were given BYR(6.6 g/kg per day and 26.6 g/kg per day respectively) every night. All the treatment courses were 30 days. The organ index was calculated. Western Blot was used to detect the expression of vimentin, claudin-11 and transferrin(TF) in testis. The serum levels of inhibin B(INHB) were determined by ELISA. Histopathological changes in testis were observed by light microscope after HE staining and evaluated by Johnsen score. Results Compared with nomal group, epididymis index, the expression of vimentin, claudin-11 and TF in testis, the level of INHB and Johnsen score significantly decreased in model group(P<0.05, P<0.01). Compared with model group, the expression of vimentin, claudin-11 and TF in testis increased in WYR group and high-dose BYR group(P<0.05, P<0.01), the level of INHB and Johnsen score significantly increased in all treated groups(P<0.01). Compared with WYR group, the expression of vimentin in testic, the level of INHB and Johnsen score significantly increased in high-dose BYR group(P<0.05, P<0.01). Conclusion The high-dose BYR significantly improved the function of sertoli cells and spermatogenesis of rats with deficiency of Shen essence,and its curative effect was better than WYR.
