摘要 目的优化药典已有标准,新建薄层鉴别方法,更加全面有效地控制黄连药材的质量。方法以硅胶G板为薄层板,盐酸小檗碱为对照品,乙酸丁酯-甲醇-异丙醇-水=5∶3∶1.5∶0.6为展开剂进行展开,紫外灯365 nm下观测,对其进行方法学考察,并对不同品种黄连、黄连混淆品进行鉴别。结果新建方法可快速鉴别出黄连中6种生物碱成分,斑点在不同薄层板、不同温度、不同湿度及微调的展开剂下均清晰可见,具有较好的耐用性,同时使用该方法还可区分黄连不同品种、不同混淆品等。结论新建方法与现有药典方法相比,展开剂使用种类少、毒性小,展开斑点多,可用于黄连药材的快速检测。 Objective To improve the quality control of Rhizoma Coptidis more effectively and optimize the standard of pharamacopoeia.Methods Thin layer chromatography was used to identify with silica gel-G plate,which was observed under ultraviolet lamp at 365 nm.Berberine hydrochloride was used as the reference substance;butyl acetate-methanol-isopropanol-water was used as the developing agent(5∶3∶1.5∶0.6).Results Six alkaloid components in coptis chinensis were identified quickly by the new method,and the spots were clearly seen under different laminates,at different temperatures,humidities and fine adjustment of developing agent ratios.Conclusion Compared with the pharmacopoeial method,this method is more reproducible and durable with little toxicity,which can be used for the rapid detection of Rhizoma Coptidis.
