摘要
目的:探讨温针灸对膝骨关节炎(Knee osteoarthritis,KOA)大鼠滑膜组织肿瘤坏死因子-α(TNF-α)水平及软骨中Wnt3a、β-catenin蛋白表达的影响。方法:将48只大鼠随机分为正常组、模型组、药物组和温针灸组,每组12只。治疗2周后,比较大鼠滑膜组织TNF-α水平及软骨中Wnt3a、β-catenin蛋白表达情况。结果:模型制备前各组大鼠LequesneMG评分差异无统计学意义。模型制备后与正常组比较,模型组、药物组、温针灸组LequesneMG评分均升高(P<0.05)。治疗2周后,与正常组比较,模型组大鼠LequesneMG评分、滑膜组织TNF-α水平、软骨Wnt3a、β-catenin蛋白表达水平明显升高(P<0.05)。与模型组比较,温针灸组和药物组大鼠LequesneMG评分、滑膜组织TNF-α水平、软骨Wnt3a、β-catenin蛋白表达水平均明显降低(P<0.05)。与药物组比较,温针灸组LequesneMG评分、软骨Wnt3a、β-catenin蛋白表达水平均明显降低(P<0.05);滑膜组织TNF-α水平差异无统计学意义(P>0.05)。结论:温针灸可以降低KOA大鼠滑膜组织TNF-α水平,其机制为抑制软骨中Wnt3a、β-catenin蛋白表达,从而达到优于西药的治疗效果,值得在临床上推广应用。
Objective: To discuss the effect of warming acupuncture and moxibustion on the level of tumor necrosis factor-α(TNF-α) in synovial tissue and expression of Wnt3 a and β-catenin in cartilage of rats with knee osteoarthritis(KOA).Methods: Divided 48 cases of rats into the normal group, the model group, the medicine group and the warming acupuncture and moxibustion group randomly, 12 rats in each group. After 2 weeks of treatment, compared the level of TNF-αin synovial tissue and expression levels of Wnt3 a and β-catenin in cartilage of rats. Results:There were no significant difference being found in LequesneMG scores of rats in each group before treatment. After establishment of models,comparing with the normal group, the LequesneMG scores in the model group, the medicine group and the warming acupuncture and moxibustion group were respectively increased(P<0.05). After 2 weeks of treatment,comparing with the normal group,the LequesneMG scores,the level of TNF-αin synovial tissue and expression levels of Wnt3 a and β-catenin in cartilage of rats in the model group were significantly increased(P<0.05). Comparing with the model group, the LequesneMG scores,the level of TNF-αin synovial tissue and expression levels of Wnt3 a and β-catenin in cartilage of rats in the warming acupuncture and moxibustion group and the medicine group were significantly decreased(P<0.05). Comparing with the medicine group,the LequesneMG scores and expression levels of Wnt3 a and β-catenin in cartilage of rats in the warming acupuncture and moxibustion group were significantly decreased(P<0.05);there was no significant difference being found in the level of TNF-αin synovial tissue of rats(P>0.05). Conclusion:Warming acupuncture and moxibustion can decrease the level of TNF-αin synovial tissue of rats. Its functional mechanism is inhabiting the expression of Wnt3 a andβ-catenin in cartilage to achieve better therapeutic effect than western medicine,which is worthy of clinical promotion and application.
