摘要
目的评价基质辅助激光解吸/电离飞行时间质谱(MALDI-TOF MS)在沙门氏菌快速鉴定及同源性分析中的应用价值.方法收集本院2017年5月至2018年6月分离的沙门氏菌,采用MALDI-TOF MS鉴定,同时采用VITEK 2 Compact全自动微生物鉴定系统鉴定和血清凝集分型,比较分析MALDI-TOF MS对沙门菌的快速鉴定准确性;对其中38株鼠伤寒沙门菌分别采用MALDI-TOF MS和PFGE同源性分析,比较两者差异.结果共分离到沙门氏菌127株,阳性率为6.55%,其中鼠伤寒沙门菌55株,占总检出沙门菌43.31%;MALDI-TOF MS可将98.42%的沙门菌准确鉴定到属,2株为没有鉴定结果;血清型水平上,质谱仪未能做到对肠道沙门菌的准确分型;对沙门氏菌同源性分析与PFGE比较具有较大差异.结论MALDI-TOF MS技术可用于沙门氏菌的快速鉴定,血清分型则需借助血清凝集试验,同源性分析与PFGE具有较大差异.
Objective To evaluate the application value of matrix—assisted laser desorption/ionization time—of—flight mass spectrometry(MALDI—TOF MS)in rapid identification and homology analysis of salmonella.Methods Salmonella isolated from our hospital from May 2017 to June 2018 were identified by MALDI—TOF MS,VITEK 2 Compact automatic microbial identification system and serum agglutination typing,and the rapid identification accuracy of MALDI—TOF MS on salmonella was compared.38 strains of salmonella typhimurium were analyzed by the homology of maldi-tof MS and PFGE respectively to compare the differences between them.Results A total of 127 salmonella strains were isolated,with a positive rate of 6.55%,among which 55 strains of salmonella typhimurium were isolated,accounted for 43.31%of the total salmonella strains.MALDI—TOF MS accurately identified 98.42%of salmonella to genus,while 2 strains had no identification results.At the level of serotype,the mass spectrometer failed to accurately classify intestinal salmonella.The analysis of salmonella homology was different from that of PFGE.Conclusion The MALDI-TOF MS technique can be used for the rapid identification of salmonella,and the serum agglutination test is needed for the serological classification.The analysis of homology is quite different from PFGE.
