摘要
目的探讨油酰乙醇胺(OEA)对糖尿病小鼠脑内炎性和神经营养因子的影响。方法采用高脂高糖加STZ链脲佐菌素(STZ)诱导T2DM小鼠模型,OEA[15、30、60 mg/(kg·d)]连续腹腔注射给药6周,通过Real time-PCR及Western blot检测海马内白介素-6(IL-6)、白介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、突触成长相关蛋白-43(GAP-43)、突触素(SYN)、BDNF和NT-3的mRNA和蛋白表达水平。结果糖尿病模型小鼠海马内炎症因子IL-6、IL-1β和TNFα的表达水平下降,并显著上调海马内GAP-43、SYN、BDNF和NT3 mRNA表达水平。结论OEA具有降低糖尿病后海马内炎症,增加海马内突触相关因子和神经营养因子的作用,从而发挥神经保护作用。
Objective To investigate the effects of oleoylethanolamine(OEA)on inflammatory and neurotrophic factors in the brain of diabetic mice.Methods T2DM mouse model was induced by high-fat,high-glucose and STZ streptozotocin(STZ),OEA[15,30,60 mg/(kg·d)]was administered by intraperitoneal injection for 6 weeks,through Real time-PCR and Western blot analysis of interleukin-6(IL-6),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),synaptic growth-related protein-43(GAP-43),mRNA and protein expression levels of synaptophysin(SYN),BDNF and NT-3.Results The expression levels of inflammatory cytokines IL-6,IL-1βand TNFαin hippocampus of diabetic mice decreased,and the expression levels of GAP-43,SYN,BDNF and NT3 mRNA in hippocampus were significantly up-regulated.Conclusion OEA can reduce the inflammation in the hippocampus after diabetes,increase the synaptic related factors and neurotrophic factors in the hippocampus,and thus exert neuroprotective effects.
