摘要
目的 研究促炎微RNA(miR)-155对间充质干细胞(DMSCs)肝细胞生长因子(HGF)和前列腺素E2(PGE2)基因表达的影响。方法 采用贴壁法分离DMSCs,分别转染miR-155类似物(mimics)以及抑制物(inhibitors)用以构建高表达或低表达miR-155的DMSC细胞系,采用实时聚合酶链反应(PCR)法比较不同DMSCs HGF和PGE2的表达水平。结果 miR-155 mimics/inhibitor转染可有效地提高或降低DMSCs的miR-155表达水平,高表达miR-155促进HGF,抑制PGE2在DMSCs的表达,而低表达miR-155则抑制HGF,促进PGE2在DMSCs的表达,提示二者均直接或间接受miR-155的调控。结论 miR-155可调节DMSCs的HGF和PGE2表达水平,进而可能对DMSCs的功能产生影响,参与银屑病局部炎症反应的发生和发展。
Objective To evaluate the influence of pro-inflammatory microRNA(miR)-155 on gene expression of hepatocyte growth factor(HGF)and prostaglandin E2(PGE2)in dermal mesenchymal stem cells(DMSCs).Methods DMSCs were expanded using adherent assay;miR-155 mimics or inhibitor was transfected into DMSCs to conduct a miR-155 over-expressing or low-expressing cell model.Real-time PCR was used to detect the gene expression of HGF and PGE2.Results miR-155 mimics/inhibitor can efficiently increase or inhibit the miR-155 expression in DMSCs,respectively.Over-expression of miR-155 induce elevated HGF and declined PGE2 expression,while low-expression of miR-155 inhibit HGF and promote PGE2 expression.Conclusion miR-155 can regulate the expression of HGF and PGE2,which may further influence the biological function of DMSCs and then participate in the pathogenesis of psoriasis.
