摘要
建立了测定微球中硫酸小檗碱含量的高效液相色谱法(HPLC)。采用C18色谱柱(4.6×250 mm,5μm),以乙腈-水(45∶55)为流动相,流速1.0 mL/min,检测波长349 nm,进样量5μL,柱温30℃。硫酸小檗碱浓度在0.098~49.00μg/mL时,其峰面积与浓度线性关系良好(R 2=0.9991),无杂质干扰,稳定性高,平均回收率为98.98%。本操作方法简便,分析快速,结果准确,适用于微球中硫酸小檗碱的含量测定。
A method for the content determination of berberine sulfuric in microspheres by HPLC was established.HPLC system consisted of C18 column(5μm 4.6×250 mm),methyl alcohol-water(45∶55)as mobile phase.The flow rate was 1.0 mL/min.The detection wave length was 349 nm.The injection volume was 5μL and column temperature was 30℃.There was no interference occurred from inpurities.The calibration curve was linear in the range of 0.098~49.00μg/mL(R 2=0.9991).The average recovery was 98.98%.The method is rapid,simple and accurate for the content determination of berberine sulfuric suspo-emulsions.
