摘要
目的:观察过山蕨总黄酮对小鼠炎症及糖尿病肾病模型大鼠相关指标的影响。方法:1)60只小鼠采用二甲苯法建立小鼠炎症模型,随机分为正常对照组,模型对照组,过山蕨总黄酮高、中、低剂量组,布洛芬片组,每组10只;2)将未造模大鼠10只设为正常对照组,采用单侧肾结扎联合腹腔注射链脲佐菌素的方法建立糖尿病肾病大鼠模型,造模成功的50只大鼠分为过山蕨总黄酮高、中、低剂量组,模型对照组,阳性对照组,每组10只。各组均灌胃给予相应的溶液,1次/d,小鼠14天,大鼠连续8周。观察各组小鼠耳肿胀,大鼠24小时尿量、血糖、血尿素氮(BUN)、血肌酐(Scr)、肾脏指数、超氧化物歧化酶(SOD)、白细胞介素6(IL-6)的影响。结果:1)过山蕨总黄酮中、高剂量组,布洛芬组与模型对照组比较,小鼠耳肿胀程度明显减轻(P<0.05);2)过山蕨总黄酮中、高剂量组,阳性对照组与模型对照组比较,可显著降低大鼠血糖、24小时尿量、血肌酐水平(P<0.05),过山蕨总黄酮高剂量及阳性对照组可显著降低大鼠的肾脏指数及血清IL-6水平(P<0.05),过山蕨总黄酮高剂量可升高大鼠血清SOD水平(P<0.05)。结论:过山蕨总黄酮可减轻二甲苯所致小鼠耳肿胀程度,对糖尿病肾病模型大鼠具有治疗作用,其机制可能与减少组织的氧化应激损伤,减少炎症因子IL-6的释放有关。
Objective:To observe th effects of total flavonoids from walking fern(Camptosorus sibiricus)on the related indexes of inflammation in mice and rat model with diabetic nephropathy(DN).Methods:1)sixty mice models were prepared using dimethylbenzene method,and randomized into normal control group,model control group,high,moderate and low doses groups of total flavonoids from walking fern and ibuprofen tablets group,ten mice each group;2)ten unhandled rats were set as normal control group,unilateral renal ligation method combined with intraabdominal injection of streptozotocin(STZ)were adopted to build DN rat models,50 rat models were randomized into high,moderate and low doses groups of total flavonoids from walking fern,model control group,positive control group and normal positive group respectively,ten rats each group.Different groups were drenched with the corresponding solution,once each day,the mice for 14 days,and the rats for eight weeks consecutively,to observe its effects on ear swelling in mice,on 24 hours urinary volume,blood glucose,BUN,Scr,renal index,SOD and IL-6 in rats in different groups.Results:1)When moderate and high dosages groups of total flavonoids from walking fern,ibuprofen tablets group were compared with model control group,ear swelling in mice relieved notably(P<0.05);2)when moderate and high dosages groups of total flavonoids from walking fern,positive control group were compared with model control group,blood glucose,24 hours urinary volume and Scr level could be reduced in rats(P<0.05),in high dose of total flavonoids from walking fern and positive control group,renal indexes and IL-6 level of rats could be lowered remarkably(P<0.05),high dose of total flavonoids from walking fern could raise SOD levels(P<0.05).Conclusion:Total flavonoids from walking fern could relieve ear swelling degrees in mice caused by dimethylbenzene,it possesses therapeutic effects on DN rats,and its mechanism might be related to relieving oxidative stress injury and reducing the release of IL-6.
