摘要
本试验旨在研究饲料蛋氨酸(Met)对小规格[初始体重为(9.75±0.05)g]和大规格[初始体重为(102.60±0.14)g]斜带石斑鱼(Epinephelus coioides)生长性能、血清生化指标和肝脏酶活性的影响,探究2个规格斜带石斑鱼的Met需求量及其对营养物质的利用能力差异。通过添加晶体DL-Met,配制Met水平分别为0.70%(0.7M)、0.85%(0.85M)、1.00%(1.0M)、1.20%(1.2M)、1.40%(1.4M)、1.60%(1.6M)、1.90%(1.9M)和2.20%(2.2M)的8种等氮等脂的饲料。将同一规格斜带石斑鱼随机分为6组,每组3个重复,小规格斜带石斑鱼每个重复放养30尾鱼,分别投喂0.7M、1.0M、1.2M、1.6M、1.9M和2.2M饲料8周;大规格斜带石斑鱼每个重复放养20尾鱼,分别投喂0.7M、0.85M、1.0M、1.2M、1.4M和1.6M饲料10周。结果表明:1)小规格斜带石斑鱼各组增重率和蛋白质效率均显著高于大规格斜带石斑鱼(P<0.05)。2)小规格斜带石斑鱼0.7M组血清总蛋白、总胆固醇和甘油三酯含量显著低于大规格斜带石斑鱼(P<0.05),血清葡萄糖含量显著高于大规格斜带石斑鱼(P<0.05)。3)小规格斜带石斑鱼1.2M组肝脏谷草转氨酶和磷酸烯醇式丙酮酸羧激酶活性显著低于大规格斜带石斑鱼(P<0.05),肝脏超氧化物歧化酶、过氧化氢酶和谷丙转氨酶活性显著高于大规格斜带石斑鱼(P<0.05)。综合以上结果,以特定生长率为判据,经二次曲线模型拟合可得出,小规格和大规格斜带石斑鱼对Met的最适需要量分别为饲料的1.40%(占饲料蛋白质的3.16%)和1.07%(占饲料蛋白质的2.38%)。与大规格斜带石斑鱼相比,在饲料Met水平低时,小规格斜带石斑鱼对葡萄糖调节能力较弱,随着Met水平升高,小规格斜带石斑鱼对糖和蛋白质的利用均得到改善。
This study was conducted to determine the effects of dietary methionine(Met)level on the growth performance,serum biochemical indices and liver enzyme activity for small size[initial weight(9.75±0.05)g]and large size[initial weight(102.60±0.14)g]of Epinephelus coioides.Exploring the Met requirement and the difference in nutrient utilization ability for two sizes of Epinephelus coioides.Eight iso-nitrogenous and iso-lipid diets were prepared with DL-Met supplement at different levels,0.70%(0.7M),0.85%(0.85M),1.00%(1.0M),1.20%(1.2M),1.40%(1.4M),1.60%(1.6M),1.90%(1.9M),and 2.20%(2.2M),respectively.Epinephelus coioides of the same size were randomly divided into 6 groups with 3 replicates in each group.Each replicate of the small size Epinephelus coioides were stocked with 30 fish and fed diets of 0.7M,1.0M,1.2M,1.6M,1.9M and 2.2M for 8 weeks,respectively.Large size Epinephelus coioides were stocked with 20 fish and fed diets of 0.7M,0.85M,1.0M,1.2M,1.4M and 1.6M for 10 weeks,respectively.The results showed as follows:1)the weight gain rate and protein efficiency ratio of the small size Epinephelus coioides were significantly higher than those of the larger size Epinephelus coioides(P<0.05).2)The contents of serum total protein,total cholesterol and triglyceride in 0.7M group of small size Epinephelus coioides were significantly lower than those of large size Epinephelus coioides(P<0.05).The content of serum glucose in 0.7M group of small size Epinephelus coioides was significantly higher than that of large size Epinephelus coioides(P<0.05).3)The activities of liver glutamic-oxaloacetic transaminase and phosphoenolpyruvate carboxylase kinase in 1.2M group of small size Epinephelus coioides were significantly lower than those of large size Epinephelus coioides(P<0.05).The activities of liver superoxide dismutase,catalase and glutamic-pyruvic transaminase in 1.2M group of small size Epinephelus coioides were significantly higher than those of large size Epinephelus coioides(P<0.05).Quadratic regression analysis of th
