摘要
目的:基于网络药理学方法,初步探讨吴茱萸抗胃癌潜在的有效成分及作用机制。方法:通过建立吴茱萸药物靶点数据集、胃癌靶点数据集,构建吴茱萸对胃癌的"成分-靶点-疾病"交互网络,并通过可视化分析、网络中各节点的拓扑参数分析,对各作用靶点进行基因本体(gene ontology,GO)富集分析及京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)通路分析,预测吴茱萸抗胃癌的作用机制。结果:筛选出3个吴茱萸主要活性成分,6个共同直接作用的蛋白靶点,8条通路参与吴茱萸治疗胃癌,包括p53信号通路、甲状腺激素信号通路和恶性肿瘤中的微小RNA等。结论:本研究通过网络药理学初步揭示了吴茱萸作用于胃癌的潜在成分及其调控网络,为其后续实验研究及临床应用提供了参考。
OBJECTIVE: To preliminarily probe into the potential active components and mechanism of Wuzhuyu in the treatment of gastric cancer based on network pharmacology. METHODS: Through the establishment of target data of Wuzhuyu and gastric cancer,Ingredients-Target-Disease interactive network of Wuzhuyu in the treatment of gastric cancer was developed. Through visualization analysis and each node of topological parameters in the network,gene ontology( GO) enrichment analysis and Kyoto encyclopedia of genes and genomes( KEGG) pathway analysis were carried out for each target to predict the mechanism of Wuzhuyu in the treatment of gastric cancer. RESULTS: Three major active ingredients Wuzhuyu were screened out,six protein targets were directly affected,and eight pathways were involved in Wuzhuyu in the treatment of gastric cancer,including p53 signal pathway,thyroid hormone signal pathway and microRNA in malignant tumors. CONCLUSIONS: This study preliminarily reveals the potential ingredients of Wuzhuyu in the treatment of gastric cancer and the regulatory network through network pharmacology,which provides reference for the subsequent experimental research and clinical application.
