N-乙酰半胱氨对醛固酮诱导血管平滑肌炎症的改善作用及对Rac1/2活化和ROS生成的影响

摘要 目的探讨N-乙酰半胱氨(NAC)对醛固酮诱导血管平滑肌细胞(VSMC)炎症的改善作用及对Rac1/2活化和活性氧(ROS)生成的影响。方法将SD大鼠分离培养的VSMCs分为对照组、醛固酮组、NACⅠ组、NACⅡ组、EHT1864组,对照组不接受任何药物处理,醛固酮组采用醛固酮(10-7 mmol/L)刺激VSMC 60 min,NACⅠ组在醛固酮刺激的同时加入0.5 mmol/L的NAC,NACⅡ组在醛固酮刺激的同时加入1 mmol/L的NAC,EHT1864组在醛固酮刺激的同时加入Eht1864(RAC1/2抑制剂)10-6 mmol/L。采用荧光素增强化学发光法检测ROS生成水平,采用蛋白印迹法检测RAC1/2和p47phox蛋白,采用RT-PCR检测VSMC中肿瘤坏死因子α(TNF-α)和白细胞介素1β(IL-1β)mRNA表达水平。结果醛固酮组VSMC的RLU高于对照组,NACⅠ、NACⅡ、EHT1864组VSMC的RLU均低于醛固酮组,NACⅡ组和EHT1864组VSMC的RLU低于NACⅠ组,差异均有统计学意义(P<0.05)。结论NAC对醛固酮升高导致血管系统疾病具有保护作用,其机制可能与抑制RAC1/2和p47phox易位与活化从而减轻NOX诱导的ROX及血管炎症有关,在心血管系统疾病的治疗中具有益作用。
