摘要
纳米技术兴起为癌症检测和治疗带来了新希望,大量研究已证明增强的肿瘤组织渗透性和滞留效应是纳米抗肿瘤药物发挥药效的关键,但是此效应也会受到体内多种因素的影响而导致纳米药物递送障碍和分布不均。这种递送障碍和分布不均主要是肿瘤血管系统异常和组织间液基质紊乱造成的。详细阐述纳米药物靶向病灶过程中的多重生理病理屏障,总结肿瘤组织微环境正常化调节手段,介绍若干用于描述抗肿瘤药物体内组织递送的生理药代动力学(PBPK)模型,为纳米药物的癌症治疗提供理论参考。
Nanotechnology brings new hope for cancer detection and treatment.Numerous studies have demonstrated that enhanced permeability and retention are key points to the efficacy of anti-tumor nano-drugs.However,this effect is affected by many factors in the body,resulting in nanoparticle delivery disorders and uneven distribution.The delivery barrier and uneven distribution are mainly caused by abnormalities in tumor vascular system and disorders in interstitial fluid matrix.The multiple physiological and pathological barriers in the nanomedicine targeting lesions were elaborated,and the normalization and regulation of microenvironment in tumor tissues were summarized.Several physiological pharmacokinetic models for tissue delivery in antitumor drugs were introduced,and provided theoretical references for cancer treatment with nano agents.
