摘要
【目的】探究海参磷脂对慢性不可预知性应激(CUMS)诱导小鼠产生抑郁样行为的影响及机制。【方法】将昆明小鼠(KM)随机分为对照组、模型组和海参磷脂低、高剂量组,对照组外的三组小鼠进行连续60 d的CUMS,同时,低、高剂量组每天分别灌胃50 mg/kg和100 mg/kg海参磷脂,模型组和对照组灌胃等体积生理盐水。应激结束后进行旷野和高架十字迷宫行为学实验,酶联免疫法检测血清糖皮质激素(GC)的含量,高效液相色谱(HPLC)检测海马区的五羟色胺(5-HT)水平。【结果】与对照组相比,模型组小鼠旷野实验5 min内水平活动、垂直运动均明显减少(P<0.01),高架实验5 min内小鼠进入高架开放臂次数显著减少(P<0.05),开放臂时间显著减少(P<0.01),血清GC显著升高(P<0.01),海马区5-HT含量显著降低(P<0.05),五羟吲哚乙酸(5-HIAA)显著升高(P<0.05),5-HT/5-HIAA比值显著降低(P<0.05)。【结论】海参磷脂可改善CUMS诱导致小鼠抑郁样行为,与其抑制GC分泌和5-HT降解活性有关。
【Objective】To investigate the effect and mechanism of sea cucumber phospholipid on depression-like behavior induced by chronic unpredictable stress(CUMS)in mice.【Methods】KM mice were randomly divided into four groups:control group,CUMS group,sea cucumber phospholipid low dose group(L)and high dose group(H).The three tested groups were given different levels of continuous CUMS for 60 days.The L group and H group were given 50 and 100 mg/kg respectively of sea cucumber phospholipid daily.Then,the Open Field Test(OFT)and Elevated Plus Maze test(EPM)were used to assess depression-like behaviors.The concentration of serum glucocorticoid(GC)were detected by ELISA.High performance liquid chromatography(HPLC)was used to detect hippocampus serotonin levels.【Results】Compared with control,there is a significant decreased in the number of rearing and locomotors in OFT,the number and the time of entries into the open arms in EPM were found in CUMS group.Additionally,the concentration of serum GC and hippocampus 5-HIAA were significantly increased,while the 5-HT content in hippocampus and the ratio of 5-HT/5-HIAA were significantly decreased in the CUMS group.However,the changes induced by CUMS can be significantly improved by sea cucumber phospholipid.【Conclusion】Sea cucumber phospholipid can improve CUMS-induced depression-like behavior in mice.This may be related to the inhibition of GC secretion and 5-HT degradation.
