摘要
以藜麦为原料,在乙醇体积分数、液料比和超声时间3个单因素的基础上,利用响应面法优化超声提取β-蜕皮激素的工艺条件。采用高效液相色谱法(HPLC)检测,以β-蜕皮激素含量为考察指标。结果表明:在246 nm波长下,最佳提取工艺条件为乙醇体积分数60%、液料比14∶1、提取时间30 min,藜麦β-蜕皮激素含量为0.3791 mg/g,与模型预测值基本相符。体外抗氧化实验表明:β-蜕皮激素对ABTS+·的清除作用显著,其IC50值为3.92μg/mL,即β-蜕皮激素具有一定的抗氧化活性。
Based on the single factor of ethanol volume fraction,liquid ratio and ultrasonic time,response surface methodology was employed to optimize the ultrasound extraction ofβ-ecdysone from quinoa.The content ofβ-ecdysone was determined by high performance liquid chromatography(HPLC)and was taken as the evaluation index.The results showed that the optimal extraction conditions at the wave of 246 nm were ethanol volume fraction of 60%,liquid ratio of 14∶1 and ultrasonic time of 30 min,and theβ-ecdysone content was determined as 0.3791 mg/g,which was basically consistent with the predicted values of the model.The results of in vitro antioxidant experiment showed thatβ-ecdysone had a significant effect on scavenging ABTS+·.The value of IC50 arrived at 3.92 g/mL,representin thatβ-ecdysone had a certain antioxidant activity.
