摘要
目的评价DNA条形码技术对基层鼠疫实验室鼠种鉴定的可行性。方法2016-2018年在平川区黄鼠疫源地采集鼠类样本,采用双盲法分别对每只鼠进行形态学鉴定和DNA条形码鉴定,比较两种方法的区别。结果平川区阿拉善黄鼠疫源地共采集鼠类174只,其中松鼠科2种、仓鼠科2种、鼠科1种、跳鼠科1种、沙鼠科1种、鼢鼠科1种和兔形目鼠兔科1种;174只鼠类COI序列所构建的进化树形成9个高度支持的分支,与形态学鉴定结果一致。结论DNA条形码分子鉴定方法适合在基层鼠疫实验室推广应用,提高鼠种分类能力。
Objective To evaluate the feasibility of the identification of rodent species by DNA barcode in the monitoring laboratory of plague in grassroots.Methods From 2016 to 2018,rodents were collected from the Spermophilus alaschanicus plague foci in Pingchuan district,Gansu province.The identification of morphological identification and DNA barcode for rodent species were carried out for each rat with double-blind method to compare the differences between the two methods.Results A total of 174 rodents were collected from Spermophilus alaschanicus plague foci in Pingchuan district,including Sciuridae(2 species),Cricetidae(2 species),Muridae(1 species),Dipodidae(1 species),Gerbillidae(1 species),Spalacidae(1 species),and Ochotonidae of Lagomorpha(1 species).The molecular evolutionary tree constructed by the COI sequence of 174 mice formed 9 highly supported branches,accorded with the morphological identification.Conclusions It is suitable for popularization and application of the method of DNA barcode identification at the laboratory of plague in grassroots to improve the classification ability of rodent species.
