大球盖菇发酵菌丝体多糖提取条件优化与结构分析 被引量:13

Optimization of Extraction and Characterization of Polysaccharide From Stropharia rugosoannulata Fermentation Mycelium

摘要 以大球盖菇发酵菌丝为试材,采用响应面法Box-Behnken设计研究了大球盖菇发酵菌丝体提取多糖最佳工艺条件,对其提取工艺进行优化,对结构进行初步表征。结果表明:最佳工艺条件为液料比26∶1 mL·g^-1、提取温度82℃、提取时间3.5 h,大球盖菇菌丝体多糖提取量为15.12%。柱前衍生化高效液相色谱分析单糖组成,包含甘露糖、核糖、鼠李糖、葡萄糖、木糖、阿拉伯糖和果糖,对应摩尔比为0.76∶1.64∶0.65∶1.00∶1.24∶1.18∶0.20。红外光谱分析具有典型的糖特征吸收。该研究结果为大球盖菇液体培养菌丝体开发利用提供参考依据。 In order to study the optimum extraction conditions and preliminarily characterization of polysaccharide from fermentation mycelium of Stropharia rugosoannulata.Box-Behnken design of response surface method was employed to optimize the extraction process.The results showed that the liquid-solid ratio,extraction temperature,extraction time of optimum process conditions were 26∶1 mL·g^-1,82℃,3.5 hours,respectively.Under these conditions,the polysaccharides yield was 15.12%.The monosaccharide composition analysis by pre-column derivatization HPLC method showed that the mycelium polysaccharide was consisted of mannose,ribose,rhamnose,glucose,xylose,arabinose and fructose in a molar ratio of 0.76∶1.64∶0.65∶1.00∶1.24∶1.18∶0.20.Meanwhile,infraredspectra showed the polysaccharide had a typical characteristic absorption of sugars.The results of this study would provide a theoretical basis for the development and utilization of mycelium from liquid fermentation of Stropharia rugosoannulata.
