摘要
目的食管癌在我国具有高发病率和高死亡率的特点,找到早期筛查和诊断的方法是提高生存率的关键。蛋白质组学在肿瘤标志物的早期筛查中具有一定的优势。本研究总结蛋白质组学在食管癌早期诊断中的应用,探讨其发展方向和最新动态。方法以"蛋白质组学、食管癌和诊断"为关键词,检索PubMed以及CNKI等期刊全文数据库2009-12-31-2019-01-01发表的相关文献,共检索到英文文献169篇,中文文献44篇,对纳入文献进行信息摘录和综合归纳。纳入标准:蛋白质组学相关研究;食管癌的诊断。根据纳入标准分析文献31篇。结果食管癌患者和健康个体之间确实存在差异蛋白,目前研究人员已经通过比较食管癌组织、血清和血浆样本筛选出一些差异蛋白和组合蛋白构建诊断模型,包括膜联蛋白A1、S100A9蛋白、微管蛋白β链和α2-HS-糖蛋白等,均具有良好的灵敏度和特异度。结论食管癌蛋白质组学研究已取得一定的进展,但蛋白质组学技术仍存在自身的局限性,缺乏足够的证据应用于临床常规检测,需进一步提高检测手段,改进研究方法,充分挖掘蛋白质组学的潜能。
OBJECTIVE Esophageal cancer is a disease with high morbidity and mortality in china.In order to improve the survival rate,it is important to develop a method for early detection and diagnosis of the cancer.There is increasing evidence that proteomics is a powerful technique for discriminating pre-cancerous and cancerous biochemical changes.This review summarized clinical applications of proteomics in the early diagnosis of esophageal cancer and discussed the research progress.METHODS In full text database exploration system like PubMed and CNKI,we used "proteomics,esophageal cancer and diagnosis" as keywords to search literature from 2009-12-31 to 2019-1-1,retrieved 169 English documents and 44 Chinese articles,the information excerpted from those literatures was analyzed and summarized.Inclusion criteria:The proteomic studies;the diagnosis of esophageal cancer.Finally,31 literature were selected.RESULTS There are some differences in proteins between patients with esophageal cancer and healthy individuals.At present,researchers have screened some differentially expressed proteins and combinatorial proteins by comparing tissues,serum and plasma samples of esophageal cancer,Annexin A1,S100 A9,tubulin beta chain and alpha 2-HS-glycoprotein have good sensitivity and specificity.CONCLUSIONS Esophageal proteomics studies have made some progress,however,proteomics technology still has its own limitations and lacks sufficient evidence for clinical routine testing.It is necessary to further improve the detection methods,improve research methods and fully tap the potential of proteomics.
