湛江卵形鲳鲹致病链球菌的分离、鉴定及药敏实验 被引量:4

Isolation, Identification and Antimicrobial Susceptibility of Pathogenic Strain from Trachinotus ovatus

摘要 为了确定2017年6月至2017年9月湛江市太平镇网箱养殖卵形鲳鲹(Trachinotus ovatus)疑似发生鱼链球菌病的病原菌及其毒力特性,取病鱼的脑、肝脏、脾脏和肾脏在血平板上进行分离培养,将分离培养的病原菌对卵形鲳鲹进行人工回归感染实验,试验鱼呈现自然发病症状:游泳异常、眼球突出且浑浊、鳍条基部充血,证明分离菌为卵形鲳鲹爆发性流行病的致病菌。通过形态学观察、生化鉴定、16S rRNA序列分析,结果表明,分离的致病菌为海豚链球菌(Streptococcus iniae),暂定为中国湛江株GHZ007(GenBank:KX788919),其对卵形鲳鲹的LD50为2.42×106CFU/mL。药敏实验结果表明,分离菌株对四环素类和氨基糖苷类药物等17种试验药物敏感,对青霉素、复方新诺明等8种试验药物有耐药性。本研究将有助于为海豚链球菌病的防治提供理论基础。 For determining the virulence of pathogenic seemly causing Trachinotus ovatus infected Streptococcus in Zhanjiang city Taiping town between June 2017 to September 2017,brain,liver,spleen,and kidney were sampled and cultured in blood plate seperately from the diseased Trachinotus ovatus.After healthy Trachinotus ovatus were re-injected with isolate,exerting symptoms were similar with previous diseased fish(erratic swimming,exophthalmia,opacification of the eye and hyperemia in fin base),which demonstrated the isolate strain contributed to the explosive epidemic of Trachinotus ovatus.The test of morphology,biochemistry and 16 S rRNA sequence analysis proved that the pathogenic bacteria,entitled with GHZ007(GenBank:KX788919)temporarily,was Streptococcus iniae,with the lethal dose(LD50):2.42×10^6 CFU/m L to Trachinotus ovatus.Antimicrobial susceptibility test showed pathogenic bacteria were sensitive to 17 antibiotics including tetracycline and aminoglycosides and resistent to anothere 8 antibiotics containing penicillin and bactrim.This reaserch will contribute to the prevention and treatment of diseases caused by Streptococcus iniae.
