摘要
目的探讨结直肠癌(CRC)组织中DOC-2/DAB2相互作用蛋白(DAB2IP)、转录因子E盒结合锌指蛋白1(ZEB1)的表达水平及临床意义。方法选取124例CRC患者的CRC组织标本及其中40例患者的癌旁正常组织标本,采用免疫组织化学染色法检测CRC组织和癌旁正常组织中DAB2IP和ZEB1的表达情况,分析CRC组织中DAB2IP与ZEB1表达的相关性,并分析DAB2IP和ZEB1表达情况与CRC患者临床特征的关系。结果CRC组织中DAB2IP的阳性表达率为80.65%,低于癌旁正常组织的97.50%(P﹤0.05);CRC组织中ZEB1的阳性表达率为44.35%,高于癌旁正常组织的12.50%(P﹤0.05)。相关性分析结果显示,CRC组织中DAB2IP与ZEB1的表达呈负相关(r=-0.737,P﹤0.01)。不同性别、年龄、肿瘤部位的CRC患者CRC组织中DAB2IP和ZEB1的阳性表达率比较,差异均无统计学意义(P﹥0.05)。低分化、有淋巴结转移的CRC患者CRC组织中DAB2IP的阳性表达率均低于中高分化、无淋巴结转移的患者(P﹤0.05);低分化、有淋巴结转移的CRC患者CRC组织中ZEB1的阳性表达率均高于中高分化、无淋巴结转移的患者(P﹤0.05)。结论CRC组织中DAB2IP表达水平降低,ZEB1表达水平升高,且CRC组织中DAB2IP和ZEB1的表达呈负相关,两者均与CRC的分化程度及淋巴结转移情况有关。
Objective To explore the expression of DOC-2/DAB2 interactive protein(DAB2IP)and transcription factor zinc finger E-box binding homeobox 1(ZEB1)in colorectal cancer(CRC)tissues and their clinical significance.Method CRC tissues from 124 patients with CRC were selected and defined as the research objects,and adjacent normal tissues from 40 patients were selected as controls.Immunohistochemistry assay was used to detect the expression levels of DAB2IP and ZEB1 in CRC tissues and adjacent normal tissues.The correlation between DAB2IP and ZEB1 expression in CRC tissues was analyzed,and the relationship between the target proteins and clinical features of CRC patients was further analyzed.Result The positive rate of DAB2IP in CRC tissues was lower than that in adjacent normal tissues(80.65%vs 97.50%)(P<0.05),while the positive rate of ZEB1 was higher than that in adjacent normal tissues(44.35%vs 12.50%)(P<0.05).Spearman correlation analysis indicated that DAB2IP expression was negatively correlated with ZEB1 expression in CRC tissues(r=-0.737,P<0.01).There was no statistically significant difference of DAB2IP or ZEB1 positive rate in terms of CRC patients with different genders,ages and tumor location sites(P>0.05),while the positive rate of DAB2IP in patients with low differentiation or lymph node metastasis was lower than that in patients with moderate-tohigh differentiation or without lymph node metastasis(P<0.05).The positive rate of ZEB1 in patients with low differentiation or lymph node metastasis was higher than that in patients with moderate-to-high differentiation or without lymph node metastasis(P<0.05).Conclusion The expression level of DAB2IP was high in CRC tissues while ZEB1 showed an opposite tendency,and both indicators were related to the differentiated degree and lymph node metastasis of CRC patients.
