摘要
目的:探讨萆苓祛痛方(萆苓方)对糖尿病痛风模型大鼠PGE2、COX-2水平的影响。方法:选取健康雄性SD大鼠50只,体重(180±20)g。适应性喂养1周,随机分为正常组、模型组、萆苓方组、吲哚组、吡格列酮组。正常组予以普通饲料喂养,其余组予高脂饲料喂养。4周后以链脲佐菌素35mg/kg腹腔注射制备糖尿病模型,以血糖≥16.7mmol/L作为糖尿病成模的标准。成模后萆苓方组按10g/kg、吲哚美辛组按5mg/kg、吡格列酮组按15mg/kg给药,正常组、模型组予等量生理盐水。4天后关节腔注射5g/L尿酸钠溶液1次,诱导痛风模型。20天后腹主动脉取血测定血糖、血尿酸、PGE2、COX-2的表达水平。结果:与正常组相比,模型组PGE2、COX-2明显升高(P<0.01);给药后,萆苓组、吲哚组指标显著降低(P<0.01)。造模后,各组动物体重均低于正常组(P<0.05或P<0.01);给药后,吡格列酮组与萆苓组体重较模型组增高(P<0.05)。与正常组相比,模型组血糖、血尿酸明显升高(P<0.01),给药后,萆苓组、吲哚美辛组血糖、血尿酸显著降低(P<0.05或P<0.01)。结论:萆苓方可帮助恢复体重,降低血糖、血尿酸水平,明显抑制PGE2、COX-2高表达。
Objective:To investigate the effects of biling qutong prescription(referred to as biling prescription)on the levels of PGE2 and COX-2 in rats with diabetic gout.Methods:Fifty healthy male SD rats were selected with a weight of(180±20)g.After 1 week of adaptive feeding,those rats were randomly divided into normal group,model group,biling prescription group,indomethacin group and pioglitazone group.The normal group was fed ordinary feed,while the rest were fed high-fat feed.Four weeks later,streptozotocin 35 mg/kg was administered intraperitoneally to induce diabetes model,and blood glucose of 16.7 mmol/L was used as the standard for diabetes modeling.After moulding,the biling prescription group was given 10 g/kg,indomethacin group 5 mg/kg and pioglitazone group 15 mg/kg,while the normal group and model group were given equal saline.After 4 days,the joint cavity was injected with 5 g/L sodium uric acid solution once to induce gout model.Blood of the abdominal aorta was taken 20 days later to determine the expression levels of blood glucose,uric acid,PGE2 and COX-2.Results:After modeling,the weight of each group was lower than that of the normal group(P<0.05 or P<0.01).After the administration,the weight of pioglitazone group and biling prescription group was higher than that of the model group(P<0.05),and there was no significant change in indomethacin group(P>0.05).Compared with the normal group,PGE2 and COX-2 in the model group were significantly increased(P<0.01).After administration,the indexes of the drug group were significantly reduced(P<0.01).Compared with the normal group,blood glucose and uric acid in the model group were significantly increased(P<0.01).After administration,blood glucose and uric acid in the zhenling group and indole group were significantly decreased(P<0.05 or P<0.01).Conclusion:Biling prescription can help to recover body weight,reduce blood glucose and uric acid level,and significantly inhibit the high expression of PGE2 and COX-2.
