摘要
目的:探讨人类APOBEC3七个蛋白结构与功能的相似与不同之处,为进一步深入研究APOBEC3的功能和抗病毒药物的开发提供参考。方法:采用生物信息学方法对APOBEC3的七个蛋白进行了理化性质、亚细胞定位、跨膜结构及相互作用蛋白的预测分析。 结果:发现人类APOBEC3均为亲水性的非跨膜蛋白,除APOBEC3H主要定位于细胞核,其余APOBEC3蛋白主要定位于细胞质;APOBEC3的二级结构非常相似,只有APOBEC3Fα螺旋所占比例最高,其余APOBEC3以无规则卷曲所占比例最高;APOBEC3DE、APOBEC3F可能还参与了细胞色素C的电子传递;APOBEC3A、APOBEC3B、APOBEC3C参与DNA修饰过程,A3H参与DNA去甲基化过程,提示APOBEC3A、APOBEC3B、APOBEC3C和APOBEC3H可能参与基因突变,导致基因组的不稳定与癌症等疾病发生;APOBEC3G参与病毒的防御应答,提示APOBEC3G具有广谱抗病毒功能。结论:人类APOBEC3是具有脱氨酶活性和水解酶活性的保守蛋白家族,可参与多种生物过程,生物学功能比较复杂。
Objective:Evaluate the similarities and differences in structures and functions of seven proteins of Human APOBEC3 to provide a reference for further research on the function of APOBEC3 and the development of antiviral drugs.Methods:Bioinformatics method was used to predict the physicochemical properties, subcellular localization,transmembrane structure,and protein interacting networks of human seven APOBEC3 members.Results:It was found that human APOBEC3 were all hydrophilic non-transmembrane proteins;except APOBEC3H was importantly located in the nucleus,the other APOBEC3 proteins were mainly located in the cytoplasm;the secondary structures of APOBEC3 were very similar,only the APOBEC3Fα helix had the highest proportion,and the remaining APOBEC3 had the highest proportion of random curl;APOBEC3DE,APOBEC3F may also participate in the electron transfer of cytochrome C;APOBEC3A,APOBEC3B,and APOBEC3C participate in the DNA modification process.APOBEC3H participates in the DNA methylation process, suggesting that APOBEC3A, APOBEC3B, APOBEC3C, and APOBEC3H may participate in gene mutations, resulting in genome instability and cancer and other diseases;APOBEC3G participates in the virus′s defense response, suggesting that APOBEC3G had a broad-spectrum antivirus function.Conclusion:Human APOBEC3 is a conserved protein family with deaminase activity and hydrolase activity,which can be involved in many biological processes and have comlex biological functions.
