白茅苷抑制AKT的活化对宫颈癌细胞增殖、凋亡和线粒体膜电位的影响 被引量:3

摘要 目的:本文旨在研究白茅苷对宫颈癌细胞增殖、凋亡和线粒体膜电位的影响。方法:采用CCK-8法检测不同剂量白茅苷对宫颈癌SiHa细胞活力的影响,选用无显著毒性的0、25、50、100μmol/L剂量白茅苷将细胞随机分为四组,克隆形成法检测各组细胞增殖;流式细胞仪检测细胞凋亡;逆转录-聚合酶链反应检测Ki67、PCNA、Bcl-2/Bax mRNA水平;流式分选检测细胞线粒体膜电位的变化;蛋白免疫印迹检测AKT磷酸化情况;免疫荧光检测AKT的细胞定位情况。结果:与Imperatorin 0μmol/L组相比较,Imperatorin 50、100μmol/L组细胞增殖率显著降低(P<0.05),凋亡率显著升高(P<0.05),Ki67、PCNA、Bcl-2/Bax mRNA水平显著降低(P<0.05),线粒体膜电位明显降低,p-AKT/AKT比值显著降低(P<0.05),荧光表达显著降低(P<0.05)。结论:白茅苷通过抑制AKT的活化抑制宫颈癌SiHa细胞增殖,促进细胞凋亡,降低线粒体膜电位。 Objective:To investigate the effects of artemisinin on cervical cancer cell proliferation,apoptosis and mitochondrial membrane potential.Methods:The effects of different doses of Imperatorin on the activity of SiHa cells in cervical cancer were detected by CCK-8 assay.Cell apoptosis was detected by flow cytometry.The mRNA levels of Ki67,PCNA and Bcl-2/Bax were detected by reverse transcription-polymerase chain reaction.The changes of mitochondrial membrane potential were detected by flow separation.Phosphorylation of AKT was detected by Western blot.Cell localization of AKT was determined by immunofluorescence.Results:The results show that compared with mu Imperatorin 0μmol/L group,50,100μmol/L of cell proliferation rate significantly decreased(P<0.05),the apoptosis rate increased significantly(P<0.05),Ki67,PCNA,Bcl-2/Bax mRNA level decreased significantly(P<0.05),mitochondrial membrane potential decreased obviously,P-AKT/AKT ratio decreased significantly(P<0.05),fluorescence expression decreased significantly(P<0.05).Conclusion:Imperatorin inhibits SiHa cell proliferation,promotes apoptosis and reduces mitochondrial membrane potential by inhibiting the activation of AKT.
