基于代谢组学木蝴蝶总黄酮抗肝肿瘤作用机制研究 被引量:9

Anti-liver cancer mechanism of total flavonoids from Oroxyli Semen based on metabolomics

摘要 目的初步阐释木蝴蝶总黄酮抗肝肿瘤的作用机制,为木蝴蝶总黄酮作为抗肝癌药物的开发和临床应用提供实验依据。方法采用二乙基亚硝胺(DEN)诱发大鼠肝癌模型,随机分为空白组,模型组,阳性对照组,木蝴蝶总黄酮高、中、低剂量组,从造模第6周开始给药,每日1次,连续10周。第17周眼球取血,分离各组血浆。采用HPLC-QTOF-MS联用技术分析木蝴蝶总黄酮干预后各组大鼠血浆中代谢物的变化,利用Agilent Mass Profiler Professional 12.6软件结合相关数据库进行主成分分析(PCA)、差异代谢物分析及代谢通路分析。结果与模型组比较,木蝴蝶总黄酮干预后共有285种代谢物发生变化,其中上调的有41种,下调的有244种。主要与色氨酸代谢、丝氨酸代谢、胆汁酸合成、花生四烯酸代谢、亚油酸代谢等途径有关。结论木蝴蝶总黄酮发挥抗肝肿瘤作用可能与调节氨基酸代谢、脂肪酸代谢、胆汁酸合成等代谢途径有关。 Objective To explain the anti-liver cancer mechanism of total flavonoids from the Oroxyli Semen,and provide experimental evidence for the development and application of total flavonoids as an antihepatocarcinoma drug.Methods Rat liver cancer models induced by diethylnitrosamine(DEN)were randomly divided into a blank group,a model group,a positive control group,and 3 dose groups of total flavonoids.The intragastric administration started at the 6 th week of modeling,once daily for 10 weeks.At the 17 th week,blood was taken from the eyeball and the plasma of each group was separated.HPLC-QTOF-MS was used to analyze the changes of plasma metabolites in the rats after the intervention of total flavonoids.Agilent Mass Profiler Professional 12.6 software was used together with the relevant database for principal component analysis,differential metabolite analysis and metabolic pathway analysis.Results Compared with the model group,changes were found in 285 metabolites after the intervention of total flavonoids,of which 41 were up-regulated and 244 were down-regulated,mainly related to tryptophan metabolism,serine metabolism,bile acid synthesis,arachidonic acid metabolism,linoleic acid metabolism and other pathways.Conclusion The anti-liver tumor effect of total flavonoids from Oroxyli Semen may be related to the regulation of amino acid metabolism,fatty acid metabolism and bile acid synthesis.
