摘要
本研究为筛选禽致病性大肠杆菌pagP基因缺失后表达量变化的相关毒力基因,对禽致病性大肠杆菌AE17及pagP基因缺失株进行RNA-Seq测序,从中筛选出与毒力相关的差异基因并对其表达量的变化进行GO和KEGG分析。以|Fold-change|≥2且FDR≤0.05为条件从测序结果中筛选得到372个差异表达基因,其中339个基因表达上调,33个基因表达下调。GO功能分类结果显示差异基因主要集中在物质跨膜运输、转Class录调控以及生物膜的形成等功能注释;KEGG数据库显示,差异表达基因主要富集在鞭毛装配、双组分系统、脂多糖的生物合成以及ABC转运蛋白等通路。pagP基因缺失会引起相关毒力基因表达量的变化,根据RNA-Seq测序结果筛选出7个对APEC具有重要作用的毒力基因,为深入研究APEC的致病机理提供依据。
To screen for changes in the expression of virulence related genes of avian pathogenic Escherichia coli with pagP gene deletion,this research used RNA-Seq on avian pathogenic Escherichia coli AE17 and the pagP deletion strain to screen differentially expressed genes of relevant virulence and analysed its variable expression via GO and KEGG.Based on the conditions of|Fold-change|≥2 and FDR≤0.05,a total of 372 genes were identified in sequencing results,among them 339 up-regulated genes,33 down-regulated genes.The GO functional classification results showed that these differentially expressed genes were focused on substance transport across the membrane,transcriptional regulation,and biofilm formation process.The KEGG database showed that these differentially expressed genes were enriched in flagellar assembly,two-component system,lipopolysaccharide biosynthesis and ABC transporters signaling pathway.Deletion of pagP gene caused changes in the expression of virulence related genes.According to the results of RNA-seq,7 virulence genes that play important roles in APEC were screened,providing bases for further study of pathogenic mechanism of APEC.
