摘要
目的观察半夏泻心汤对脓毒症大鼠肠道机械屏障的保护作用,并初步探讨其相关机制。方法将30只SD大鼠随机分为对照组、脓毒症组和半夏泻心汤组。采用盲肠结扎穿孔(CLP)法制备脓毒症大鼠模型。对照组只开腹后关腹,其他操作相同。半夏泻心汤组在成模时开始灌胃中药。对照组和脓毒症组在成模时灌胃生理盐水。成模后72 h处死大鼠,留取血清和肠道组织标本进行组织病理学评价,检测血清内毒素、D-乳酸、二胺氧化酶(DAO)、肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)含量以及组织TNF-α、IL-10的基因表达。结果半夏泻心汤组大鼠肠道组织病理损伤情况较脓毒症组明显减轻。半夏泻心汤组血清内毒素、D-乳酸、DAO、TNF-α含量均较脓毒症组显著减少,而IL-10含量较脓毒症组显著增加(P<0.05)。半夏泻心汤组肠道组织TNF-α的基因表达水平较脓毒症显著降低,而IL-10的基因表达水平较脓毒症显著增加(均P<0.05)。结论半夏泻心汤能减轻炎症反应,对脓毒症大鼠肠道机械屏障具有保护作用,其机制可能是通过上调IL-10表达和抑制TNF-α表达实现的。
Objective:To investigate the protective effect of Banxia Xiexin Decoction on intestinal mechanical barrier in sepsis rats and explore its probable mechanism.Methods:30 Sprague-Dawley(SD)rats were randomly divided into control group,sepsis group and Banxia Xiexin Decoction group.Sepsis model was established by cecal ligation and puncture(CLP).The control group only closed the abdomen after laparotomy.The Banxia Xiexin Decoction group began to infuse Chinese herbal medicine at the same time as the model was established.The control group and the sepsis group were infused with saline.The rats were killed 72 hours after the establishment of the model,and the serum and intestinal tissue samples were collected.Histopathological evaluation,the contents of endotoxin,D-lactic acid,DAO,TNF-α,IL-10 in serum and the gene expression of TNF-αand IL-10 in tissues were detected.Results:The pathological damage of intestinal tissue in Banxia Xiexin Decoction group was significantly less than that in sepsis group.The levels of endotoxin,D-lactic acid,DAO and TNF-α in the Pinelliae Decoction group were significantly lower than those in the sepsis group,while the levels of IL-10 were significantly higher than those in the sepsis group.The gene expression level of TNF-α in intestinal tissue of Banxia Xiexin Decoction group was significantly lower than that of sepsis group,while the gene expression level of IL-10 was significantly higher than that of sepsis group.Conclusion:Banxia Xiexin Decoction,plays a role in the regulation of inflammatory response and alleviate the inflammatory reactions,has a protective effect on intestinal mechanical barrier of sepsis rats.The up-regulated IL-10 expression and down-regulated TNF-α expression might be served as its potential mechanism.
