摘要
为筛选白鲜愈伤组织培养的适宜培养基,试验以延边地区野生白鲜种子为材料,研究了MS培养基中植物生长调节剂种类及其浓度对愈伤组织诱导和生长以及愈伤组织中白鲜碱、黄柏酮和梣酮积累的影响。结果表明:在MS+2.0 mg/L NAA+2.0 mg/L 6-BA以及MS+2.0 mg/L 2,4-D+2.0 mg/L 6-BA培养基中,愈伤组织诱导率及生长较好;在MS+2.0 mg/L 2,4-D+1.0 mg/L 6-BA培养的愈伤组织中,白鲜碱和梣酮的有效积累量最高,在MS+2.0 mg/L 2,4-D+2.0 mg/L 6-BA培养的愈伤组织中,黄柏酮的有效积累量最高。
In orther to screen the suitable medium for callus culture,seeds of wild Dictamnus dasycarpus growing in Yanbian area were used as materials in this experiment,the effects of plant growth regulators and their concentrations in MS medium on inducing and growth of callus,and accumulation of dictamnine,obacunone and fraxinellone in callus were investigated.The results showed that the callus grew well in the MS+2.0 mg/L NAA+2.0 mg/L 6-BA,and MS+2.0 mg/L 2,4-D+2.0 mg/L 6-BA medium.The effective accumulation of dictamnine and fraxinellone in callus on the MS+2.0 mg/L 2,4-D+1.0 mg/L 6-BA medium,and obacunone in callus on the MS+2.0 mg/L 2,4-D+2.0 mg/L 6-BA medium were the highest.
