摘要 药代动力学主要研究机体对药物处置的动态变化,哺乳动物组织和器官的生理功能呈现明显的昼夜节律,因此药物的药代动力学也可能产生相似的24 h振荡变化,从而影响药效与安全性。对于窄治疗窗或者高变异的药物来说,探索昼夜节律对其药动学的影响很可能具有重要的临床意义。本文就昼夜节律对药物代谢动力学影响的研究进展以及在药物临床试验中的探索与应用进行综述。 Pharmacokinetics is a study aiming to investigate the dynamic changes of drug disposition in the body,while the organisms and tissues of mammalian are demonstrated to be highly organized by circadian rhythms.Therefore,it is not surprising that many aspects of pharmacokinetics also oscillate according to the same 24-hour clocks which lead to the variety on efficacy and safety of drugs.In terms of drugs with narrow therapeutic windows or high variability,exploring the effects of circadian rhythms on their pharmacokinetics is probably of highly significant clinical implications.This review focuses on the advance progress of circadian rhythms’effects on pharmacokinetics of drug,and its roles in the exploration and application of clinical trials.
