我国六个地方鸭品种的群体遗传结构及遗传分化 被引量:2

Population genetic structure and genetic differentiation of six local duck breeds in China

摘要 【目的】了解中国部分地方鸭品种的群体遗传结构和遗传分化.【方法】使用荧光标记的微卫星引物和ABI-3730XL型DNA测序仪器,对我国6个地方鸭品种(180只)在16个微卫星位点上的等位基因进行STR分型测序,利用Structure和Fstat软件对群体结构和遗传分化进行分析.【结果】16个微卫星位点共检测到141个等位基因,具有高度多态性;F-统计量结果显示群体间存在显著的遗传分化;Structure程序成功构建出了6个鸭品种的遗传分化结构图;Structure程序构建的Neighbor Joining(NJ)图将‘润州凤头白鸭’‘于田麻鸭’聚为一类,‘北京鸭’与‘樱桃谷鸭’‘吉安红毛鸭’聚为一类.【结论】通过16个多态性较好的微卫星位点对中国6个地方鸭品种进行了遗传结构性和遗传分化研究,为获取我国地方鸭品种间的遗传信息和遗传关系提供更准确的依据. 【Objective】To understand the genetic structure and genetic differentiation of local duck species in China.【Method】In this study,the fluorescent-labeled microsatellite primers were used for STR typing of alleles in 16 microsatellite loci in 6 duck cultivars(180 heads)preserved in China by ABI-3730XL DNA sequencing platform.Population structure and genetic differentiation were analyzed using Structure and Fstat software.【Result】A total of 141 alleles were detected in 16 microsatellite loci with high polymorphism.The F-statistics showed significant genetic differentiation among populations.The Structure program successfully constructed the genetic differentiation structure of 6 duck breeds.The Neighbor Joining(NJ)map constructed by the Structure program showed that‘Runzhou White Crested Duck’and‘Yutian Ma Duck’were clustered together,and‘Beijing Duck’‘Cherry Valley Duck’and‘Ji'an Red-haired Duck’were clustered together.【Conclusion】In this study,16 microsatellite loci with abundant polymorphism were used to study the genetic structure and genetic differentiation of 6 local ducks in China,which provided more accurate basis for obtaining the genetic information and genetic relationship among local ducks in China.
