雪峰虫草液体培养产三萜及其抗菌作用的初步研究 被引量:1

Preliminary Study on Production and Antimicrobial Activity of Triterpenoids from Xuefeng Congcao(Ophiocordyceps xuefengensis) by Liquid Cell Culture

摘要 目的:通过液体培养体系培养雪峰虫草菌体合成三萜化合物,并检测其三萜提取物对大肠杆菌等5种菌的体外抑菌作用。方法:液体培养法和杯碟法。结果:适宜雪峰虫草细胞生长和三萜产物合成的最佳条件是:起始pH 6.0,摇床转速160 r/min,培养温度28℃,培养时间8 d,接种量15%,装液量100 mL/250 mL。在此条件下,雪峰虫草三萜产量可达197.28 mg/L,虫草细胞生物量可达10.87 g/L。大肠杆菌和枯草芽孢杆菌对该三萜提取物较为敏感,其MIC均为20 g/L,产黄青霉最不敏感,MIC为80 g/L。结论:通过液体培养体系培养雪峰虫草菌体合成三萜化合物具有可行性,所得三萜类化合物具有较好的抑菌作用。 Objective:To study the production and antimicrobial activity of triterpenoids from Ophiocordyceps xuefengensis by liquid cell culture.Methods:Liquid culture and cylinder-plate method.Results:The optimum conditions for the cell growth and triterpenoids synthesis were as follows:initial pH 6.0,rotating speed 160 r/min,culture temperature 28℃,culture time 8 days,inoculum volume 15%,liquid volume 100 mL/250 mL.Under these conditions,the yield of triterpenoids and the cell biomass of P.xuefengensis reached 197.28 mg/L and 10.87 g/L respectively.The crude triterpenoid extract had obvious inhibitory effect on the growth of Escherichia coli and Bacilllus subtitis,however,the smallest inhibitory effect on Penicillium chrysogenum was observed.The minimal inhibitory concentration(MIC)of the triterpenoid extract to E.coli,B.subtitis and P.chrysogenum was 20,20 and 80 g/L.Conclusion:It is feasible to produce triterpenoids from P.xuefengensis cultured in liquid culture system,and the triterpenoids obtained had good antimicrobial activity.
