摘要
目的:探讨麝香通心滴丸对糖尿病心肌病的保护作用及其与心肌锚蛋白Ankrd1蛋白表达的相关性。方法:共选取40只小鼠,以随机数字表法分为3组,即正常对照组(正常组)、糖尿病心肌病组(模型组)、麝香通心滴丸组(滴丸组),腹腔注射STZ复制糖尿病心肌病模型,麝香通心滴丸组给予麝香通心滴丸50 mg/(kg·d)灌胃,连续给药4周;彩超测定各组小鼠心功能指标,HE染色观察各组小鼠的心肌病理改变,蛋白免疫印迹法检测各组小鼠心肌组织Ankrd1蛋白的表达。结果:滴丸组相较于模型组,LVDd、LVDs、FS、LVEF心功能指标均明显改善(均P<0.05);HE染色显示,滴丸组心肌细胞肥厚及心肌细胞排练紊乱相较于模型组明显减轻;Western Blot结果可见滴丸组Ankrd1的表达相较于DM组明显降低(P<0.05)。结论:麝香通心滴丸可有效减轻糖尿病小鼠的心肌病变,其机制可能与下调Ankrd1蛋白的表达相关。
Objective:To research the protective effect of Shexiang Tongxin Dropping Pills on diabetic cardiomyopathy by regulating the expression of Ankrd1 protein.Methods:40 male C57 mice were randomly divided into three groups:control group(Control),diabetic cardiomyopathy group(model)and Shexiang Tongxin Dropping Pill group(MSM).Diabetic cardiomyopathy model was established by intraperitoneal injection of STZ,and Shexiang Tongxin Dropping Pill group was given Shexiang Tongxin Dropping Pill on the basis of diabetic cardiomyopathy group for 4 weeks,the cardiac function indexes of mice in each group were detected by color Doppler ultrasonography,the cardiac structure of mice in each group was determined by HE staining,and the expression of Ankrd1 protein in myocardial tissue of mice in each group was detected by Western blotting.Results:Compared with model group,LVDd,LVDs,FS and LVEF in MSM group were significantly improved(all P<0.05),HE staining showed that the cardiac hypertrophy and cardiac dysfunction caused by diabetic cardiomyopathy in MSM group were effectively alleviated;the expression of Ankrd1 was determined by Western Blot was significantly decreased in MSM group(P<0.05).Conclusion:Shexiang Tongxin Dropping Pills effectively alleviated myocardial lesions in diabetic mice,and its mechanism may be related to down-regulation of the expression of Ankrd1 protein.
