早搏灵对乌头碱诱导心律失常大鼠心肌组织KCNJ2表达的影响 被引量:1

Effects of Zaoboling(早搏灵) on KCNJ2 Expression in Cardiomyocytes of Rats with Tachyarrhythmia Induced by Aconitine

摘要 目的:通过观察早搏灵对乌头碱诱导的心律失常大鼠的心肌KCNJ2(内向整流钾通道J亚家族成员2)基因表达的影响,探讨复方中药早搏灵抗心律失常的作用机理。方法:选用健康大鼠40只随机分4组,正常对照组、模型组、早搏灵组和胺碘酮;连续给药1周后,除正常组外其余各组大鼠舌静脉注射乌头碱造模,同时描记大鼠心电图变化,采用实时荧光定量PCR技术观察心肌组织KCNJ2基因表达的变化。结果:与模型组比较,早搏灵组和胺碘酮组早搏、室速出现时间均明显延迟(P<0.05),早搏灵组和胺碘酮组比较无明显差异(P>0.05);早搏灵组室颤出现时间与模型组比较无明显差异(P>0.05)。与正常组比较,模型组大鼠心肌组织KCNJ2基因表达明显降低(P<0.05),早搏灵组、胺碘酮组KCNJ2基因表达与模型组比较明显提高(P<0.05),早搏灵组和胺碘酮组比较无明显差异(P>0.05)。结论:早搏灵胶囊可延迟乌头碱诱导的大鼠早搏、室速出现时间,上调心肌组织KCNJ2基因表达。 Objective:To observe the effects of Zaoboling on the expression of KCNJ2 gene in cardiomyocytes of rats with aconitine induced arrhythmia,and to explore the mechanism of Zaoboling in antiarrhythmia.Methods:40 healthy rats were randomly divided into four groups:normal,model,Zaoboling and Amiodarone groups.After one week of continuous administration,aconitine was injected into the tongue vein of model group,Amiodarone group and Zaoboling group,and the changes of ECG were recorded at the same time.The changes of KCNJ2 gene expression in myocardial tissue were observed by real-time fluorescence quantitative PCR.Results:To compare with model group,the occurrence time of premature beats and ventricular tachycardia in Zaoboling and Amiodarone groups were delayed significantly(P<0.05),there was no significant difference between the two medicine groups on the the occurrence time of premature beats and ventricular tachycardia(P>0.05);there was no difference between Zaoboling group and model group on the occurrence time of ventricular fibrillation(P>0.05).The KCNJ2 gene expression in rats’cardiomyocytes of model group was reduced to compare with normal group(P<0.05),the KCNJ2 gene expression was strengthened in Zaoboling and Amiodarone groups,that was higher than that of model group(P<0.05),there was no significant difference between the two medicine groups on KCNJ2 gene expression(P>0.05).Conclusion:Zaoboling capsule can delay the occurrence of premature beats and ventricular tachycardia in rats induced by aconitine,and up-regulate the expression of KCNJ2 gene in cardiomyocytes.
