摘要
目的研究有氧运动对小鼠主动脉自噬相关因子表达及自噬活化的影响。方法将8周龄C57BL/6雄性小鼠40只随机分为安静对照组(n=20)与有氧运动组(n=20),其中10只用于Real-Time PCR实验,10只用于Western-blot实验,有氧运动组小鼠进行6周的游泳运动(60 min/d,5 d/周),对照组小鼠自由活动。6周后取小鼠胸腹主动脉,采用Real-Time PCR方法检测自噬相关因子ULK1、Beclin1、LC3B mRNA水平;Western-blot方法检测主动脉LC3B蛋白中LC3B-II/LC3B-Ⅰ比值及p62蛋白水平。结果与对照组相比,有氧运动组小鼠主动脉ULK1、Beclin1、LC3B mRNA水平明显升高(P<0.05);有氧运动组小鼠主动脉LC3B-II/LC3B-Ⅰ比值增大(P<0.05),p62蛋白降解增加(P<0.05)。结论有氧运动能够使主动脉ULK1、Beclin1、LC3B自噬相关因子表达增强,进而增强主动脉自噬活性。
Objective To investigate the effect of aerobic exercise on the levels of autophagy-related markers and Autophagy activation in mouse aorta. Methods Twenty C57 BL/6 male mice aged 8 weeks were randomly divided into sedentary control group and aerobic exercise group. The aerobic exercise group were subjected to swimming for 6 weeks, 60 min/d, 5 days a week. The Thoracic and abdominal aorta of mice were isolated and detected after 6 weeks. Real-Time PCR was adopted to measure the mRNA levels of autophagy-related markers, including ULK1, Beclin1 and LC3 B. Western-blot was applied to measure the expression level of autophagy-related protein including the ratio of LC3 B-II/LC3 B-Ⅰ and p62. Results Compared with the control group, in the aerobic exercise group, the mRNA levels of ULK1, Beclin1 and LC3 B and the ratio of LC3 B-II/LC3 B-Ⅰ were significantly increased(P < 0.05), the protein degradation of p62 was significantly increased(P < 0.05). Conclusion Aerobic exercise can increase the expression of ULK1, Beclin1 and LC3 B in the aorta, and then enhance the aortic autophagy activity.
