摘要
采集海南6个山兰米酒酒厂的酒曲样品,经基因组DNA提取,分别对细菌16SrDNA和真菌ITS区域进行PCR扩增及MiSeq测序,同时进行物种注释、丰富度聚类以及多样性分析。试验结果表明,酒曲中真菌主要有根霉属(Rhizopus)、假丝酵母属(Candida)、Bullera、Simplicillium、曲霉属(Aspergillus)、Gibberella,其中根霉属是酒曲中真菌的优势菌属;细菌主要有片球菌(Pediococcus)、肠杆菌属(Enterobacter)、魏斯氏菌Weissella、Klebsiella等,其中片球菌含量最多。基于6个酒厂的微生物群落的Alpha和Beta多样性分析,酒厂E的酒曲细菌群落的物种丰富度及均匀度都较高,酒厂C的酒曲细菌物种丰富度较低,6个酒厂的酒曲样品细菌和真菌存在显著差异。
he rice wine starter samples were collected from six wineries in Hainan province,after genomic DNAextraction,the bacterial 16 S rDNA and fungal ITS regions were amplified and Illumina MiSeq sequencing was performed.The species annotation and diversity analysis were analyzed.The fungi inShanlan Rice Wine are mainly Rhizopus,Candida,Bullera,Simplicillium,Aspergillus,Gibberella,Rhizopusis the dominant genus of fungi;the bacteria mainly include Pediococcus,Enterobacter,Weissella,Klebsiella,etc.,of which the highest content of Pediococcus.Based on the Alpha and Beta diversity analysis of the microbial communities of the six wineries,The species richness and uniformity of the bacterial community in winery E were higher,and the richness of distiller’s bacterial species in winery C was lower.there were differences in the diversity of bacterium and fungi communities of shanlan rice wine starter collected from different wineries.
