姜黄素对幼鼠海马乙醇敏感受体NMDAR1重塑的影响 被引量:1

摘要 目的:探讨姜黄素对酒精中毒幼鼠学习、记忆能力的影响及其机制。方法:将雄性3周龄SD大鼠(50~70 g)按简单随机抽样的方法分成酒精组、对照组、治疗组、NaOH组,每组8只。酒精组用52%乙醇溶液灌胃[10 mL/(kg·d)]制作酒精中毒模型;对照组用等量生理盐水灌胃,余同酒精组;治疗组在酒精组基础上加用姜黄素NaOH溶液[100 mg/(kg·d)]进行腹腔注射;NaOH组仅用等体积、同浓度的NaOH进行腹腔注射。7 d后采用Morris水迷宫观察每组幼鼠的学习、记忆功能;13 d后采用免疫组化观察大脑海马冠状切面切片海马区N-甲基-D-天冬氨酸受体1(NMDAR1)表达情况。结果:与对照组相比,酒精组在目标象限停留时间显著缩短,海马区NMDAR1阳性细胞累积光密度(IOD)值明显降低(均P<0.05);与酒精组相比,治疗组在目标象限停留时间显著延长(P<0.05),海马区NMDAR1阳性细胞IOD值明显升高(均P<0.05)。结论:姜黄素对酒精中毒幼鼠学习记忆功能损害有改善作用,其机制可能与增加幼鼠海马区NMDAR1表达有关。 Objective:To investigate the effect and mechanisms of curcumin on learning and memory abilities in alcoholic young rats.Methods:Thirty-two male 3-week-old SD rats(50-70 g)were randomly divided into alcohol group,control group,treatment group and NaOH group,with 8 rats in each group.Alcoholism model was made by intragastric administration of 52%ethanol solution[10 mL/(kg·d)].The rats in the control group were given 52%ethanol solution via gavage and the same amount of saline.The rats in the treatment group were given 52%ethanol solution and then intraperitoneal injection of curcumin NaOH solution[100 mg/(kg·d)],and rats in the NaOH group were given intraperitoneal injection with the same concentration of NaOH only.After 7 days,Morris water maze was used to observe learning and memory function of each group rats.After 13 days,the expression of NMDAR1 in coronal section of hippocampus was observed by immunohistochemistry.Results:Compared with the control group,the time spent in the target quadrant in the alcohol group was significantly reduced,and IOD value of NMDAR1 positive cells in the hippocampus was decreased(P<0.05).Compared with the alcohol group,the time spent in the target quadrant in the treatment group was significantly increased,and IOD value of NMDAR1 positive cells in the hippocampus was increased(P<0.05).Conclusion:Curcumin could improve the learning and memory abilities of young rats with alcoholism,and the mechanism might be related to the increase of NMDAR1 expression in the hippocampus.
