摘要
目的比较五味子总多糖(SCP)及总木脂素(SCL)对酒精及对乙酰氨基酚(APAP)诱导的两种不同小鼠急性肝损伤模型的保护作用。方法将ICR小鼠随机分为5组,每组20只:即正常对照组(CON,蒸馏水),模型组(MOD,蒸馏水),多糖组(SCP,100mg/kg),木脂素组(SCL,100mg/kg)和多糖+木脂素组(SCP+SCL,SCP50mg/kg,SCL50mg/kg),每天灌胃1次,每次10ml/kg,连续14天。末次灌胃1小时后,除CON组外其他各组小鼠分别灌胃50%乙醇溶液(10ml/kg,一次)用于制成酒精肝损伤模型;或腹腔注射250mg/kgAPAP生理盐水溶液(10ml/kg,一次)用于制成APAP肝损伤模型,而CON组给予同剂量的蒸馏水。12~16小时后处死小鼠,测量肝脏指数,血清中谷丙转氨酶(ALT)和谷草转氨酶(AST)的水平。结果SCP、SCL及SCP+SCL组均可显著降低酒精和APAP所致模型小鼠肝脏指数及血清ALT、AST水平(P<0.05或P<0.01),而在APAP模型中,SCP组对ALT和AST水平的降低效果显著优于SCL及SCP+SCL组。结论SCP及SCL对酒精和APAP诱导的肝损伤均有显著的保护作用,但对于APAP所致肝损伤,SCP的效果最佳。
Objective To compare total Schisandra chinensis polysaccharide(SCP)and total Schisandra chinensis lignans(SCL)on two different acute liver injury models induced by alcohol and acetaminophen(APAP)in mice.Methods ICR mice were randomly divided into 5 groups,20 in each:normal control group(CON,distilled water),model group(MOD,distilled water),(SCP,100mg/kg),total lignan group(SCL,100mg/kg)and total polysaccharide+total lignan group(SCP+SCL,SCP 50mg/kg,SCL 50mg/kg),oral gavage,once daily,10ml/kg at a time,and for 14 consecutive days.One hour after the last administration,mice in each group except the CON group were administered with oral 50%ethanol solution(10ml/kg,once)to make an alcohol liver injury model;or intraperitoneal 250mg/kg APAP physiological saline solution(10ml/kg,once)to make an APAP liver injury model,and the CON group was given the same dose of distilled water orally or intraperitoneally,respectively.Mice were sacrificed after 12~16 hours,liver index,serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)levels were measured.Results SCP,SCL and SCP+SCL groups could significantly reduce liver index,serum ALT and AST levels in model mice caused by alcohol and APAP(P<0.05 or P<0.01),while in the APAP model,the effects of SCP group on reducing ALT and AST levels were significantly better than SCL and SCP+SCL groups.Conclusion SCP and SCL had significant protective effects on alcohol and APAP-induced liver damages,but SCP had the best effect on liver damage caused by APAP.
