摘要
应用胶体金免疫层析技术,通过对标记抗体的pH值、抗体浓度以及重悬液的条件优化,进行西马特罗胶体金快速检测卡的研制。结果表明,其灵敏度达到0.1ppb。该检测卡简单、快速、灵敏度、特异性、稳定性、重复性好,与盐酸克伦特罗、沙丁胺醇、莱克多巴胺等常见的瘦肉精均无交叉反应。检测86份猪组织样品与ELISA结果符合率为91.7%。结果表明,研制的西马特罗胶体金快速检测卡,可应用于基层养殖、屠宰、市场环节动物及动物产品的的现场快速检测。
The colloidal gold immunochromatography was applied, and pH value, concentration of antibody and condition of resuspending of marked antibody were optimized, and quick detection card for cimaterol colloidal gold was developed. The results showed that the sensitivity reached 0.1 ppb. The detection card was simple and rapid, and sensitivity, specificity, stability and repeatability were good, and there was no cross reaction with common lean meat essence such as clenbuterol, salbutamol and ractopamine. The 86 pig tissue samples were detected, and coincidence rate with ELISA was 91.7%. The results showed that the quick detection card for cimaterol colloidal gold developed could be applied to rapid detection of primary culture, slaughtering, market segment animals and animal products in field.
