苦碟子对大鼠肾缺血再灌注损伤的预防作用

摘要 目的探讨苦碟子对大鼠肾缺血再灌注损伤(RIRI)的预防作用。方法将30只SD大鼠分为假手术组(等体积0. 9%氯化钠溶液)、模型组(等体积0. 9%氯化钠溶液)和苦碟子组(4 m L/kg),各10只。以无创动脉夹夹闭大鼠双侧肾动脉,建立大鼠肾脏缺血再灌注损伤模型。苏木素-伊红(HE)染色,观察大鼠肾组织形态学变化,检测大鼠血清肌酐(SCr)和尿素氮(BUN)水平,以及肾匀浆白细胞介素6(IL-6)、超氧化物歧化酶(SOD)和丙二醛(MDA)水平;采用Western Blot法检测肾组织中核转录因子-κB(NF-κB)蛋白表达水平。结果模型组大鼠肾小管上皮细胞明显受累,肾小管管腔扩张伴管型形成,肾小球有炎性细胞浸润;苦碟子组肾小管损伤程度明显轻于模型组。与假手术组比较,模型组大鼠血清SCr和BUN,肾匀浆IL-6,MDA,NF-κB蛋白表达水平均显著升高,肾匀浆SOD水平显著降低(P <0. 05);与模型组比较,苦碟子组大鼠血清SCr和BUN,肾匀浆IL-6,MDA,NF-κB蛋白表达水平均显著降低,肾匀浆SOD水平显著升高(P <0. 05)。结论大鼠肾缺血再灌注前使用苦碟子能缓解大鼠RIRI,其机制可能与抗氧化应激并下调NF-κB信号转导通路从而缓解炎性损伤有关。 Objective To evaluate the preventive effect of Kudiezi on renal ischemia reperfusion injury( RIRI) in rats. Methods A total of 30 male rats were randomly divided into three groups: sham group( 9% sodium chloride solution for equal volume),kidney ischemia-reperfusion group( KIR group,9% sodium chloride solution for equal volume),and Kudiezi group( KDZ group,4 m L/kg),10 rats in each group. The renal ischemia reperfusion injury model was established by non-invasive arterial clipping of bilateral renal arteries in rats. Hematoxylin and eosin( HE) staining was used to observe the pathological changes of renal tissue,and the levels of serum creatinine( SCr) and blood urea nitrogen( BUN) as well as the content of interleukin 6( IL-6),superoxide dismutase( SOD) and malondialdehyde( MDA) in renal tissue were detected. Western blot was performed to determine the expression level of NF-κB protein in the renal tissue. Results In the KIR group,the epithelial cells of renal tubules was obviously involved;meanwhile,the renal tubule cavity was dilated with tubulogenesis,and the glomerulus was infiltrated by inflammatory cells. The damage degree of renal tubules in the KDZ group was significantly lower than that in the KIR group. Compared with the sham group,the SCr and BUN levels,the content of IL-6 and MDA,the expression level of NF-κB protein were significantly increased,while the content of SOD was significantly decreased in the KIR group( P < 0. 05). Compared with the KIR group,the SCr and BUN levels,the content of IL-6 and MDA,the expression level of NF-κB protein were significantly increased,while the content of SOD was significantly decreased in the KDZ group( P < 0. 05).Conclusion Prevention with Kudiezi before reperfusion can relieve RIRI effectively. The mechanism may correlate with the anti-oxidative stress of Kudiezi and the downregulation of NF-κB signaling pathway to alleviate the inflammation injury.
