酒鬼酒发酵车间空气源细菌系统发育多样性研究 被引量:4

Phylogenetic Diversity of Airborne Bacteria in Fermentation Workshops of Jiugui Distillery

摘要 本研究采用空气自然沉降法和纯培养法从酒鬼酒发酵车间收集分离空气中可培养细菌(含放线菌),运用基于16S rRNA基因序列的系统发育分析法对菌株进行多样性研究。用添加10%酒鬼酒酒醅浸汁的营养琼脂、麦芽膏-酵母膏和高氏一号培养基,从酒鬼酒发酵车间分离纯化到127株细菌菌株。根据形态观察和部分生理生化实验去冗余,选取72株具有代表性的菌株,对其进行基于16S r RNA基因序列的系统发育多样性分析。结果表明,这些分离菌株具有较高的类群多样性,分属于细菌域的4个门(Actinobacteria,Deinococcus-Thermus,Firmicutes,Proteobacteria)的17个科(Aurantimonadaceae,Bacillaceae,Bacillales FamilyⅫ,Corynebacteriaceae,Deinococcaceae,Intrasporangiaceae,Microbacteriaceae,Moraxellaceae,Nocardiaceae,Planococcaceae,Propionibacteriaceae,Pseudomonadaceae,Rhizobiaceae,Rhodobacteraceae,Sphingomonadaceae,Staphylococcaceae,Xanthomonadaceae)的24个属(Acinetobacter,Agrococcus,Arsenicicoccus,Arthrobacter,Aurantimonas,Bacillus,Corynebacterium,Deinococcus,Exiguobacterium,Janibacter,Kocuria,Luteococcus,Microbacterium,Micrococcus,Paracoccus,Pararhodobacter,Pseudomonas,Rhodococcus,Shinella,Sphingomonas,Sporosarcina,Staphylococcus,Virgibacillus,Xanthomonas)。优势类群为Actinobacteria门(34株,47.22%),其中Micrococcaceae为优势科,含15株菌株;其次是Proteobacteria门(24株,33.33%)和Firmicutes门(11株,15.28%),还分离到3株属于分类名称尚未确定且目前根据《伯杰氏细菌手册》暂称为异常球菌-栖热菌门(Deinococcus-Thermus)的细菌。这些菌株可分为53个物种,大部分菌株与其系统发育关系最密切的已知物种的典型菌株之间存在一定的遗传差异(16S rRNA基因序列相似性为96.37%~99.88%)。其中至少有5株(JSM 2215019、JSM 2215034、JSM2215039、JSM 2215042和JSM 2215109)可能代表潜在的新的分类单元(potential new taxa),它们可能分别代表Sphingomonas,Corynebacterium,Arthrobacter,Paracoccus和Deinoc In this study,the airborne bacteria in the fermentation workshops of Jiugui Distillery were collected by using natural precipitation method,and then isolated by using culture-dependent method.After the purificaiton,the biodiversity of the isolated bacteria was investigated by using phylogenetic analysis based on 16S rRNA sequences.A total of 127 bacteria strains were isolated on a number of media(nutrient agar,ISP 2 and Coates’mediumⅠagar)supplemented with 10%extract of the fermenting grains of Jiugui liquor.Based on the results of morphological characteristics and physiological/biochemical tests,72 strains were selected for further study.Phylogenetic analysis results showed that,these 72 strains belonged to 24 genera(Acinetobacter,Agrococcus,Arsenicicoccus,Arthrobacter,Aurantimonas,Bacillus,Corynebacterium,Deinococcus,Exiguobacterium,Janibacter,Kocuria,Luteococcus,Microbacterium,Micrococcus,Paracoccus,Pararhodobacter,Pseudomonas,Rhodococcus,Shinella,Sphingomonas,Sporosarcina,Staphylococcus,Virgibacillus,Xanthomonas)of 17 families(Aurantimonadaceae,Bacillaceae,Bacillales FamilyⅫ,Corynebacteriaceae,Deinococcaceae,Intrasporangiaceae,Microbacteriaceae,Moraxellaceae,Nocardiaceae,Planococcaceae,Propionibacteriaceae,Pseudomonadaceae,Rhizobiaceae,Rhodobacteraceae,Sphingomonadaceae,Staphylococcaceae,Xanthomonadaceae)in 4 major phylogenetic groups(phylum)(Actinobacteria,Deinococcus-Thermus,Firmicutes,Proteobacteria).The dominant phylum was Actinobacteria(34 strains,47.22%),followed by Proteobacteria(24 strains,33.33%)and Firmicutes(11 strains,15.28%).The other 3 strains were members of the Deinococcus-Thermus phylogenetic group.The 72 strains could be classified as 53 genomic species,and the majority of the strains,to some extent,exhibited genetic discrepancy with their relatives(96.37%to 99.88%sequence similarity).At least 5 strains(JSM 2215019,JSM 2215034,JSM 2215039,JSM 2215042 and JSM 2215109)should represent potential new species of the genera Arthrobacter,Corynebacterium,Deinococcus,Paracoccus and Sphingomonas
