甜瓜黄绿叶色性状的遗传分析及其初步定位 被引量:5

摘要 为了研究叶色突变对甜瓜光合性能的影响并定位调控叶色的基因,对厚皮甜瓜黄绿叶色材料M68和叶色正常材料M465进行叶绿素荧光动力参数测定。结果显示,与M465相比,M68暗适应后的最小荧光值Fo、最大荧光值Fm及可变荧光值Fv均显著降低,而Fv/Fm比值显著升高,表明M68叶绿素含量下降导致PSII电子传递的最大潜力及光化学反应能力显著下降,同时PSII最大光量子效率增强。利用M68和M465构建BC1群体,对控制甜瓜黄绿叶色的基因进行遗传规律分析和基因定位研究。结果表明,甜瓜黄绿叶色性状是由1对隐性单基因控制的,命名为yellow-green leaf(ygl)基因。利用分离群体分组分析法(bulked segregate analysis,BSA)对BC1群体中的绿色与黄绿叶色2个混池进行简单重复序列(simple sequence repeats,SSR)标记筛选,初步将ygl基因定位在甜瓜第11染色体上。ygl基因与两侧最近的SSR标记分别相距0.6 cM(CmSSR25149)和0.3 cM(CmSSR25196),物理距离1180 kb。 In order to study the effect of leaf color mutation on the photosynthetic performance in Melon and map leaf color controling genes,the chlorophyll fluorescence kinetic parameters of M68 with yellow-green leaf and M465 with normal green leaf were determined and compared.The results showed that,compared with M465,the minimal fluorescence yield(Fo),the maximal fluorescence yield(Fm)and the variable fluorescence yield(Fv)of the dark-adapted state in M68 were significantly lower than those in M465,while the Fv/Fm ratio in M68 was significantly higher than that in M465,indicating that the chlorophyll content decrease in M68 had led to a significant decrease in its maximum potential of PSII electron transfer and photochemical reaction ability,while the biggest PSII light quantum efficiency is enhanced.M68 and M465 were further used to construct BC1 population to conduct research on the genetic analysis and mapping of the genes controlling the yellow-green leaf color of the Melon.Genetic analysis revealed that the yellow-green leaf traits in M68 were controlled by a pair of single recessive genes,which was named yellow-green leaf(ygl)gene.Bulked segregate analysis(BSA)was used to screen the green and yellow green color pools in the BC1 population with SSR marker,and the gene was initially mapped on chromosome 11.On this basis,the ygl gene were preliminarily mapped at a point with a physical distance of 0.6 cM(from CmSSR25149)and 0.3 cM(from CmSSR25196),the nearest SSR markers on both sides,with physical distance of 1180 kb between the two SSR markers.
