摘要
为探索Ghrelin在幼犬体内的表达及分布情况,采用实时荧光定量PCR(RT-qPCR)和免疫组化染色对Ghrelin在消化器官的表达情况进行了定量及定位检测。RT-qPCR结果显示,在检测的12个消化器官中,Ghrelin相对表达量差异显著。胃的表达量最高,显著高于其他组织,其次是舌、十二指肠、肝、回肠、空肠、食管,而下颌腺、胰腺、盲肠、直肠的表达量较低,结肠的表达量最低。免疫组化结果显示,Ghrelin主要分布在舌、食管、胃和肠管的黏膜层和肌层,黏膜下层也可见阳性细胞分布;Ghrelin在肝和胰腺主要分布在肝细胞胞质、胰腺腺泡细胞的胞核和胞质,下颌腺阳性细胞主要分布在闰管、纹状管和导管的管壁。结果提示,Ghrelin可能对幼犬消化管的消化、吸收和分泌功能及消化腺的分泌活动具有重要的调控作用。
To quantify the expression and detect the distribution of Ghrelin in digestive organs of puppies,real-time quantitative PCR(RT-q PCR) and immunohistochemical staining techniques were used.The RT-q PCR results showed that the difference of the relative expression of Ghrelin among the 12 digestive organs tested was significant.The highest expression of Ghrelin was found in the stomach,which was significantly higher than that observed in the other tissues.Then,the higher expression of Ghrelin was successively found in the tongue,duodenum,liver,ileum,jejunum and esophagus;while the expression was lower in the mandibular gland,pancreas,cecum and rectum.The expression in the colon was the lowest.The immunohistochemical results showed that Ghrelin was mainly distributed in the mucosal and muscular layers of the tongue,esophagus,stomach and intestine;besides,positive cells were also observed in the submucosa.While in the liver and pancreas,Ghrelin was mainly distributed in the cytoplasm of hepatocytes,and the nucleus and cytoplasm of pancreatic acinar cells.In the mandibular glands,positive cells were mainly distributed in the wall of the intercalated duct,striated duct and catheter.The results suggested that ghrelin might play important roles in regulating functions of digestive,absorptive and secretory in digestive tracts,and secretory activities in digestive glands in puppies.
