摘要 为分离并鉴定A群猪轮状病毒(PoRV),本研究通过MA-104细胞分离培养、间接免疫荧光鉴定、RT-PCR鉴定以及序列分析对一份疑似Po RV感染的小肠样本进行了病毒的分离与鉴定。结果显示,分离到一株Po RV,其基因型为G9-P[13]-I5-R1-C1-M1-A8-NI-T7-E1-H1,命名为RVA/Pig-tc/CHN/SWU-1C/2018/G9P[13]。经序列分析显示,该病毒的VP7、VP4、VP6、NSP1、NSP2、NSP4和NSP5基因与Po RV相似性最高,达92.9%~98.3%,而VP1~VP3、NSP3基因与人源A群轮状病毒(GAR)相似性最高,达94.1%~97.3%,表明该病毒为一株人猪重配的Po RV。经软件重组分析显示,该病毒的VP2基因在626 bp处与人源GAR R1207株发生重组,进一步证实该病毒为人猪重组病毒株。本研究首次在中国大陆报道分离鉴定到一株猪源人猪重配的G9P[13]型Po RV,为后续对其致病性研究奠定了基础。 To isolate and identify the porcine group A rotavirus(PoRV),a small intestine sample suspected of PoRV infection was isolated using MA-104 cells and was further identified by indirect immunofluorescence assay,RT-PCR and sequence analysis.The results showed that the virus isolate was PoRV,which genome constellation was G9-P[13]-I5-R1-C1-M1-A8-NI-T7-E1-H1,and named as RVA/Pig-tc/CHN/SWU-1 C/2018/G9 P[13].Sequence analysis showed that the homology of VP7,VP4,VP6,NSP1,NSP2,NSP4 and NSP5 genes of the isolate had the highest identities with PoRV,with 92.9%-98.3%similarity,while VP1-VP3 and NSP3 genes had the highest homologies with human group A rotavirus(GAR),with 94.1%-97.3%similarity.The above results indicated that this rotavirus strain was a reassortant between human and porcine rotavirus.Recombination analysis showed that the VP2 gene of the virus recombined with human GAR R1207 strain at 626 bp,which further confirmed that the virus was a human and porcine reassortant strain.In addition,this is the first time isolated and identified a reassortant G9 P[13]PoRV between human and porcine strains in China's Mainland,which lays a foundation for further pathogenicity research.
