摘要
探究不同浓度茉莉酸甲酯诱导下马尾松(Pinus masssoniana Lamb.)的防御反应及其时序变化规律,为研究外界胁迫诱导马尾松的抗性机理奠定基础。分别用浓度为0.1、1.0、10.0 mmol/L的茉莉酸甲酯溶液处理2年生马尾松幼苗,并于处理后6、15、30、45 d采集不同处理组马尾松茎干、树梢,制作石蜡切片和徒手切片,用显微镜观察采集马尾松的各类型树脂道的面积及诱导产生的创伤性树脂道数量。茉莉酸甲酯处理马尾松后,茎的组织解剖学结构表现出创伤性树脂道的形成、皮层树脂道腔隙增大和多酚薄壁细胞的膨胀与增加等现象。创伤性树脂道的发生发育可分为原始细胞阶段、胞间隙形成及腔道扩大阶段和树脂道成熟阶段。不同浓度茉莉酸甲酯处理后,马尾松创伤性树脂道的形成数量具有明显差异。茉莉酸甲酯可以诱导马尾松产生组织解剖结构的防御应答,通过研究分析其应答规律,可为深入探索马尾松诱导性的防御机理提供理论依据。
This study was conducted to study the temporal sequence of secondary defense in Pinus massoniana induced by different concentration of methyl jasmonate(MeJA)to provide a theoretical basis for the integrated management of P.massoniana pests.Two-year-old P.massoniana seedlings were treated with MeJA at concentrations of 0.1,1 and 10 mmol/L,respectively.Stems and leaders of every single seedling in different treatments were collected at 6,15,30 and 45-d post treatment,respectively.Paraffin and hand-sliced sections were applied in measuring the resin duct size and counting their number.Traumatic resin duct formation,cortical resin duct cavity expansion as well as the increase of both number and size of polyphenol thin-wall cells were found after MeJA treatment.Traumatic resin duct formation could be divided into following stages:primitive cell,interstitial formation and lumen expansion and eventually resin duct maturation.Number of newly formed traumatic resin ducts in P.massoniana significantly varied when treated with MeJA of different concentrations.In P.massoniana,the sequential differentiation of traumatic resin ducts can be induced and influenced by varied MeJA solution.
