摘要
目的探讨葛根素注射液联合雌二醇注射液对大鼠骨质疏松性骨折愈合的影响。方法选取SPF级健康雌性成年SD大鼠70只,随机分为对照组14只及造模组56只。造模组用去卵巢手术及切断股骨建立骨质疏松性骨折模型,对照组切断股骨造成非骨质疏松性骨折。将模型大鼠随机分为模型组、实验A组、实验B组和实验C组,每组14只。对照组和模型组均给予0.9%NaCl 1mL·kg^-1;实验A组给予葛根素注射液50 mg·kg^-1;实验B组给予苯甲酸雌二醇注射液80μg·kg^-1;实验C组给予葛根素注射液30 mg·kg^-1和苯甲酸雌二醇注射液60μg·kg^-1。5组大鼠均给予皮下注射,qd,连续给药8周。比较5组大鼠的骨密度(BMD)值,股骨骨痂组织骨保护素(OPG)、核因子κB受体活化因子(RANK)和RANK配体(RANKL)蛋白表达水平。结果治疗后,实验A组、实验B组、实验C组、模型组和对照组的右股骨BMD分别为(0.29±0.04),(0.34±0.04),(0.38±0.05),(0.25±0.03)和(0.42±0.05)g·cm-2,股骨骨痂组织OPG蛋白相对表达量分别为(0.57±0.07),(0.81±0.08),(1.06±0.11),(0.38±0.05)和(1.29±0.16),RANK蛋白相对表达量分别为(0.89±0.08),(0.67±0.05),(0.42±0.06),(1.01±0.09)和(0.25±0.03),RANKL蛋白相对表达量分别为(0.94±0.12),(0.80±0.09),(0.49±0.07),(1.12±0.13)和(0.31±0.04),模型组的上述指标与对照组和实验A、B、C组比较,差异均有统计学意义(均P<0.05)。结论葛根素注射液联合雌二醇注射液能显著改善骨质疏松性骨折大鼠的骨密度,其作用可能与调节OPG/RANK/RANKL信号通路活性有关。
Objective To analyze the effects of puerarin injection combined with estradiol injection on healing of osteoporotic fracture in rats.Methods A total of 70 SPF female adult rats were randomly divided into control group with 14 rats and model building group with 56 rats.The model building group was established osteoporotic fracture model by ovariectomy and femoral transection,rats in the control group was established non-osteoporotic fracture by femoral transection.The model rats were randomly divided into model group,experimental A group,experimental B group and experimental C group with 14 rats per group.The osteoporotic fracture model was established by ovariectomy and cutting off the femur in the model building group,and the non-osteoporotic fracturewas induced by cutting off the femur in the control group.Control and model groups were given 0.9%NaCl 1 mL·kg^-1.Experimental A group was treated with puerarin injection 50 mg·kg^-1.Experimental B group received estradiol benzoate injection 80μg·kg^-1.Experimental C group was given puerarin injection 30 mg·kg^-1 combined with estradiol benzoate injection 60μg·kg^-1.Five groups were given subcutaneous injection once a day for 8 weeks.The bone mineral density(BMD),the protein expression levels of osteoprotegerin(OPG),nuclear factor kappa B receptor activating factor(RANK)and RANK ligand(RANKL)infemoral callus tissue were compared.Results After treatment,the main indexes of experimental A group,experimental B group,experimental C group,model group and control groups were compared:the BMD of right femurs were(0.29±0.04),(0.34±0.04),(0.38±0.05),(0.25±0.03)and(0.42±0.05)g·cm-2,the OPG protein relative expression levels were(0.57±0.07),(0.81±0.08),(1.06±0.11),(0.38±0.05)and(1.29±0.16),RANK protein relative expression levels were(0.89±0.08),(0.67±0.05),(0.42±0.06),(1.01±0.09)and(0.25±0.03),RANKL protein relative expression levels were(0.94±0.12),(0.80±0.09),(0.49±0.07),(1.12±0.13)and(0.31±0.04).There were significant differences between the expe
