摘要
目的探讨PDLSCs细胞膜片与纳米级B-TCP复合叠层三维结构对牙槽骨缺损的相关修复及治疗作用。方法分离及培养牙周膜干细胞,制备牙周膜干细胞生物膜片,构建PDLSCs细胞膜片与纳米级B-TCP复合叠层三维结构模型,将其植入新西兰兔缺损骨模型内,于术后第4周、第8周、第12周处死新西兰兔并分离牙周组织,通过形态学分析评判修复效果。结果 WB结果示,Runx2、ALP蛋白的表达量比对照组分别增加9.2倍和4.3倍;移植后第8周,苏木精-伊红染色示PDLSCs细胞膜片与纳米级B-TCP复合叠层三维复合结构组新骨形成的百分比显著高于对照组及空白对照组(P<0.05);免疫组化染色示构建PDLSCs细胞膜片与纳米级B-TCP复合叠层三维复合结构组成骨相关标志物OCN、ALP、Runx2蛋白表达高于对照组及空白对照组(P<0.05)。结论牙周膜干细胞膜片与B-TCP三维叠层结构复合结构可促进兔牙槽骨缺损部位的修复及再生,为临床牙槽骨缺损的治疗提供一种新的思路,有广阔的应用前景。
Objective To investigate the repair and treatment effects of PDLSCs membrane and nano-scale B-TCP composite laminated three-dimensional structure for alveolar bone defects.Methods The periodontal ligament stem cells were isolated and cultured to prepare the biomembranes of periodontal ligament stem cells.The composite laminated three-dimensional structure model of PDLSCs membrane and nano-scale B-TCP was constructed and implanted into the New Zealand rabbit defect bone model.The New Zealand rabbits were sacrificed and the periodontal tissues were isolated in the 4 th week,8 th week and 12 th week after surgery.The repair effect was evaluated by morphological analysis.Results WB results showed that the expression levels of Runx2 protein and ALP protein were 9.2 times and4.3 times higher than those in the control group.In the 8 th week after transplantation,hematoxylin-eosin staining showed that the percentage of new bone formation in the PDLSCs membrane and nano-scale B-TCP composite laminated three-dimensional structure group was significantly higher than that in the control group and the blank control group(P<0.05).Immunohistochemical staining showed that expression levels of the ossification related markers such as OCN,ALP and Runx2 in the PDLSCs membrane and nano-scale B-TCP composite laminated three-dimensional structure group were higher than those in the control group and the blank control group(P<0.05).Conclusion Periodontal ligament stem cells membrane and nano-scale B-TCP composite laminated three-dimensional structure can promote the repair and regeneration of rabbit periodontal bone defect,which provides a new idea for the treatment of clinical alveolar bone defect and has a wide application prospect.
